内皮祖细胞移植治疗糖尿病大鼠下肢动脉缺血的动脉造影改变
Angiogenesis observed by angiography in endothelial progenitor cells transplantation to hindlimb ischemia rat model with diabetes
【摘要】 目的对比观察内皮祖细胞(EPCs)和骨髓单个核细胞移植治疗糖尿病大鼠下肢动脉缺血的动脉变化。方法将雄性Wistar大鼠30只诱导成糖尿病模型,其中25只成模,成模鼠结扎左下肢股动脉制造下肢缺血模型,此后随机分成3组,分别给于生理盐水(A组,n=7),内皮祖细胞(B组,n=9),骨髓单个核细胞(C组,n=9),沿股动脉走向、多点肌肉注射。继续喂养28 d后行下肢动脉造影检测下肢血流。结果动脉造影结果显示EPCs移植治疗组缺血侧下肢动脉显影血管数多于对照组(P<0.05);EPCs移植治疗组血管数多于骨髓间充质干细胞移植治疗组,但无统计学差异。结论 EPCs移植治疗对糖尿病大鼠下肢缺血效果明确。
【Abstract】 Objective To compare the effects of transplantation of endothelial progenitor cells(EPCs) and bone marrow mononuclear cell on hind limb ischemia in diabetic rat model.Methods The male STZ-induced diabetes Wistar rats model were randomly divided into 3 groups: Sodium Chloride control group(A group),EPCs transplantation group(B group) and bone marrow mononuclear cell group(C group),each group had 9 mouse.Rats with hind limb ischemia were treated by transplantation of ex vivo expanded EPCs and bone marrow mononuclear cells,the effects were assessed by angiography.Results Angiography showed that angiogenesis index was significantly higher in EPCs transplantation rats with diabetes versus control at 28th day.The effect of EPCs transplantation was slightly better than that of the bone marrow transplantation,but was not statistically significant.Conclusion EPC transplantation is an effective therapeutic method for treatment of hind limb ischemia in diabetic rats model.
