血管变异对糖尿病大鼠急性下肢缺血模型的影响及动脉造影的评估意义
Influence of variation angiogenesis in acute hind limb ischemia diabetic rat model by using angiography method
【摘要】 目的通过动脉造影法对糖尿病鼠下肢急性缺血模型进行评估,探讨血管变异对糖尿病急性下肢缺血模型构建的影响。方法 30只雄性Wistar鼠,经高脂喂养后腹腔注射STZ(40 mg/kg)诱发糖尿病模型,血糖达16.8 mmol/L后,再手术结扎股动脉,造成左下肢急性缺血模型,喂养28 d后用动脉造影法测定下肢血流。结果 83.3%大鼠(25只)空腹血糖水平达16.8 mmol/L,结扎后1~7 d,大体观察下肢显著性缺血(P<0.05),28d大体观察下肢形态基本恢复到术前水平,缺血侧下肢肌肉较对侧略萎缩,但行动自如;动脉造影显示,28 d后来自于髂内动脉的动脉分支明显增粗,其远端血管代偿性增加,供应膝关节及以下区域。结论大鼠下肢有来自于髂内动脉的分支参与供血,此变异对缺血模型的建立有影响;使用动脉造影法可以发现此类变异,并准确评估糖尿病下肢缺血及血管的代偿性改变。
【Abstract】 Objective To evaluate the model of diabetic rats′ acute hind limb ischemia through the method of angiography,and to explore the influence of variation angiogenesis for contributing the model.Methods Thrity male Wistar rats after fed with high-lipid food were intra-peritoneal injected streptozocin,streptozotocin(STZ,40 mg/kg) to evoke diabetic model.Femoral artery of the diabetic rats model were ligated to cause acute hind limb ischemia.The model rats were fed 28 days,the angiogenesis was observed by angiography method on the 28 days.Results There were 25 rats(83.3%) with the fasting blood glucose more than 16.8 mmol/L,the hind limb was obviously ischemia(P<0.05) observed by eye on first 7 days after ligation,and the ischemia almost recovered to condition of preoperative after 28 days,muscle of ischemic hind limb slightly atrophy compared with the other side,but movement was convenient.After 28 days,arteries branch from internal iliac artery thickened evidently,blood vessels of the distal end raised compensatory,providing the area of knee joint and the blow.Conclusion There are branches from internal iliac artery to participate the blood-supply,the variation have influences on establishing the ischemia model;by using angiography can discovery this kind of variation,and accurately evaluate the diabetic hind limb ischemia and the compensatory changing of blood vessels.
