谷氨酸干预体外大鼠星形胶质细胞MRP1表达

【摘要】 目的在体外无神经元作用下,观察谷氨酸对星形胶质细胞表达多药耐药相关蛋白1(MRP1)的影响,探讨谷氨酸与多药耐药现象的关系。方法取新生SD大鼠脑,分离纯化星形胶质细胞,适当传代后加用含谷氨酸500μmol/L、1000μmol/L的培养基干预3天、7天、10天,通过免疫细胞化学检测细胞MRP1的表达。结果与对照组比较,谷氨酸干预后MRP1表达水平升高,3天表达水平稍升高,7天表达水平升高明显并可持续至10天余,低浓度组与高浓度组作用无明显差异。结论 MRP1在星形胶质细胞中仅少量表达;谷氨酸环境培养可促进体外大鼠星形胶质细胞MRP1的表达。

【Abstract】 Objective To observe effects of glutamate on levels of multidrug resistance-associated protein 1(MRP1) expressed by rat astrocytes in vitro without the impact of neurons and explore the relationship between multidrug resistance phenomenon and glutamate.MethodsAstrocytes from newborn SD rats were purified,cultured and incubated with medium of 500μmol/L,1000μmol/L glutamate for 3d,7d and 10d,respectively.The expression of MRP1 was investigated with immunocytochemistry.ResultsCompared with the negative control group,increased expression of the MRP1 was detected.The amount of the protein increased slightly in 3 day,significantly 7 days and maintained to more than 10 days.There was no dose-dependent manner between the up-regulation of MRP1 expression level and the concentrations of glutamate.ConclusionsGlutamate may affect the overexpression of MRP1 in astrocytes,thus play an important role in the drug-resistant mechanism of refractory epilepsy.
