应用流式细胞仪检测结核分枝杆菌对左氧氟沙星的敏感性
STUDY ON SENSITIVITY OF MYCOBACTERIUM TUBERCULOSIS TO LEVOFLOXACIN DETECTED BY FLOW CYTOMETRY
【摘要】 目的利用流式细胞仪检测结核分枝杆菌对左氧氟沙星的敏感性。方法利用结核分枝杆菌水解FDA的特点,用FDA标记经过左氧氟沙星作用后的标准菌株H37RV和临床分离菌株,利用流式细胞仪检测标记了FDA的菌株的荧光强度,并与比例法结果进行比较。结果 40株结核分枝杆菌的流式细胞仪所得药敏试验结果与比例法基本一致(p>0.05)。结论流式细胞仪检测结核分枝杆菌药敏实验的方法极大地缩短了获得药敏结果的时间,可以考虑作为常规项目开展。
【Abstract】 Objective To detect drug sensitivity of Mycobacterium tuberculosis to levofloxacin by flow cytometry. Methods Based on the ability of viable M.tuberculosis organism hydrolyzing flourescein diacetate(FDA),we detect the sensitivity of levofloxacin of H37RV type strain and 40 clinical isolated Mycobacteria strains incubated with levofloxacin by flow cytometry using FDA.The result was compared with that of proportion method results. Results The result obtained by flow cytometry and proportion method did not change significantly.(p>0.05). Conclusion Flow cytometry susceptibility testing is a rapid and convenient method for Mycobacterium tuberculosis.
