新型双链季铵盐对金黄色葡萄球菌损伤作用的电镜观察

【摘要】 [目的]以金黄色葡萄球菌为对象,研究双链季铵盐N,N,N′,N′-四甲基-N,N′-双十二烷基-2-丁烯-1,4-二铵(简称1,4-DBeQA)的杀菌机制。[方法]采用载体定量杀菌和扫描电镜观察的方法进行实验观察。[结果]用含量100mg/L的(1,4-DBeQA)消毒液作用10min,对金黄色葡萄球菌的杀灭率达到100%。通过扫描电镜的观察发现,25mg/L的(1,4-DBeQA)消毒液作用20min,金黄色葡萄球菌菌体仍保有较完好的外部形态,但菌体表面粗糙,有少数菌体胞壁出现皱褶。100mg/L的(1,4-DBeQA)消毒液作用20min,金黄色葡萄球菌的细胞壁有显著皱褶和破裂,已观察到细菌表面有许多颗粒状物出现,并有小泡和小球形成。[结论]从双链季铵盐(1,4-DBeQA)杀菌机制的角度看来,细菌的致死与细胞质的凝集和胞内物质的渗漏有关。

【Abstract】 [Objective]To study the germicidal mechanism of 1,4-bis(N,N-dimethyl-N-dodecyl ammonium bromide)-2-butylene(1,4-DBeQA)with staphylococcus aureus as the experimental object.[Methods]Carrier quantitative bactericidal test and scanning electronmicroscope observation method were used to carry out laboratory observation.[Results]The disinfectant solution containing(1,4-DBeQA)100mg/L with a 10 min contact time killing rate 100% of staphylococcus aureus,Scanning electronmicroscopic observation found that after exposure to 25mg/L(1,4-DBeQA)for 20min staphylococcus aureus thalli still had intact external shape.The rough of bacterial surface and a few bacterial thalli showed obvious folds.After exposure to 100mg/L(1,4-DBeQA)for 20min,the cell wall of staphylococcus aureus showed obvious folds and rupture.It has been observed that the bacterial surface has a many pellet substance,small bubble and spherical body.[Conclusion]The germicidal mechanism of bisquats(1,4-DBeQA)significant correlated with the agglutination of cytoplasm and leakage of intracellular substance and death of bacteria.
