延边朝鲜族人群LDLR基因多态性与原发性高血压相关性研究

【摘要】 [目的]探讨延边朝鲜族人群低密度脂蛋白受体(LDLR)基因第12外显子C1773T多态性与原发性高血压的相关性。[方法]采用聚合酶链反应-单链构象多态(PCR-SSCP)技术,结合DNA直接测序方法,对122例高血压患者和146例血压正常人进行LDLR基因型检测;同时也对他们的10余项相关临床生化指标进行测定,采用ANOVA、χ2检验等方法进行统计学分析。[结果]高血压组等位基因C的频率为89.34%,对照组等位基因C的频率为80.47%,两者之间比较差异有统计学意义(P﹤0.05);患者组的体重指数、体脂肪率明显高于对照组,而血浆脂联素水平低于对照组(P﹤0.05)。[结论]延边朝鲜族人群LDLR基因第12外显子C1773T多态性与原发性高血压的发生有相关性,等位基因C可能是延边朝鲜族原发性高血压发生的一个危险基因;高体重指数、体脂肪率和低血浆脂联素水平可能是延边朝鲜族人群高血压发生的重要危险因素。

【Abstract】 [Objective]To study the relationship between the C1773T polymorphism in exon 12 of the low density lipoprotein receptor(LDLR)gene and essential hypertension(EH)in Yanbian Korean-Chinese populations.[Methods]We enrolled 122 EH patients and 146 healthy individuals.More than 10 clinical biochemical indicators were measured.PCR-SSCP and DNA sequencing were used to test LDLR genotypes.ANOVA,χ2 test were used for analysis of data.[Results]The frequency of C allele in EH group(89.34%)was obviously higher than that in normal group(80.47%),and there was a statistical significance(P﹤0.05).The body mass index(BMI),body fat rate in EH group were higher than in control group,but the level of plasma adiponectin in EH group was less than in control group(P﹤0.05).[Conclusion]There is significantly associated between the C1773T polymorphism in exon 12 of the LDLR gene and EH in Yanbian Korean-Chinese populations.The allele C could be considered as a risk gene in the progression of EH in Yanbian Korean-Chinese.The higher BMI and body fat rate,and the lower plasma adiponectin are important risk factors with EH in Yanbian Korean-Chinese populations.
