α-氯代醇对大鼠的亚慢性经口毒性试验研究

【摘要】 [目的]研究α-氯代醇对大鼠的主要亚慢性毒性作用,确定毒作用靶器官及其毒作用剂量。[方法]参照GB15670-1995《农药登记毒理学试验方法》中大鼠亚慢性毒性经口毒性试验方法进行,设0,15,30,75mg/kg4个剂量组。[结果]与对照组比较,雄性高剂量组(75mg/kg)体重增长明显减慢(P﹤0.01),两性别高剂量组血常规中的血红蛋白含量和RBC计数均较对照组降低(P﹤0.05),两性别中、高剂量组肾脏脏器系数有明显改变(P﹤0.05或P﹤0.01),雄性中、高剂量组睾丸及两性别中、高剂量肾脏存在明显病理改变,雄性各剂量组精子计数、精子活动度明显降低(P﹤0.05或P﹤0.01)。[结论]在本试验条件下,连续摄入30mg/kg以上≥80%α-氯代醇90d对SD大鼠肾脏有显著损伤,连续摄入15mg/kg≥80%α-氯代醇90d对雄性SD大鼠精子活动度及精子计数就有显著影响,连续摄入30mg/kg以上≥80%α-氯代醇90d对雄性SD大鼠生殖系统有显著病理损伤。

【Abstract】 [Objective]To study on the subchronic oral toxicity of α-chlorohydrin on rats and to obtain its NOAEL and targeted organs.[Methods]According to the subchronic oral toxicity testing method listed in the Toxicological Testing Methods for Pesticide Registration(GB15670- 1995),4 doses groups(0,15,30,75mg/kg)were set up.[Results]The body weights of male rats in high dose group(75 mg/kg)were significantly less than those of the groups(P﹤0.01),decreased values for haemoglobin concentration and erythrocyte count were found for both male and female treated rats in high dose group,the ratios of kidney weights in 30,75 mg/kg group were higher than that in the control group(P﹤0.05 or P﹤0.01),testes of male rats and kidney of both male and female rats in 30,75mg/kg group had obvious pathological damage,decreased values for sperm count and sperm motion were found for all tested male rats(P﹤0.05 or P﹤0.01).[Conclusion]The 90 days subchronic oral toxicity test shows that the exposure of 30mg/kg/d for ≥80%α-chlorohydrin has obvious damage effect on the kidney of both male and female SD rats,the exposure of 15mg/kg/d for ≥80% α-chlorohydrin has obvious effect on sperm count and sperm motion of male rats in all tested groups. Moreover,the exposure of 30mg/kg/d for ≥80% α-chlorohydrin has remarkable damage effect on reproductive system of male SD rats.
