埃柯19型病毒的鉴定及VP1区基因特征分析
ANALYSIS OF THE CHARACTERISTICS OF ECHOVIRUS 19 ISOLATION AND SEQUENCING OF VP1 REGION
【摘要】 [目的]研究河南省埃柯19型病毒分离株VP1区基因特征。[方法]从2009年手足口病例标本中分离病毒,提取RNA并逆转录,应用VPl区特异型引物扩增VPl区全长基因片段并进行序列测定、系统发生树构建和核苷酸同源性分析。[结果]从2009年河南省手足口病人标本中分离出的肠道病毒,其中一株VP1区扩增成功,VPl全基因序列测定和同源性比较分析表明,该分离株为埃柯19型肠道病毒,且与山东省2009年分离株同源性最高,达到97.3%。[结论]河南省手足口病原谱中存在埃柯19型病毒,并与山东分离株属同一分支。
【Abstract】 [Objective] To study the gene characterization of echovirus 19 virus strains isolated in Henan province.[Methods]Viruses were isolated from clinical specimens of hand foot and mouth disease(HFMD)in 2009 of Henan province.The positive isolates were identified by reverse transcriptase PCR(RT-PCR)with specific primer pair for VP1 genes.Complete VP1 gene sequences were determined and compared with other isolates available from GeneBank by homogeneity and phylogenetic tree analyses.[Results]Enterovirus were isolated from clinical specimens and one of them was amplified using VP1 genes primers.The sequencing result indicated that Henan strain belong to Echoviurs 19,and the highest nucleotide homogeneity of Henan strain with Shandong strain was 97.3%.[Conclusion]Echovirus 19 exists in Henan HFMD pathogen spectrum,and it belongs to the same branch with Shangdong strain.
