金雀异黄素对U251MG的抑制作用及对癌基因表达的影响
THE EFFECTS OF GENISTEIN ON PROLIFERATION AND ONCOGENE EXPRESSION OF HUMAN GLIOMA CELL LINE U251MG
【摘要】 [目的]探讨金雀异黄素(Genistein)对胶质瘤细胞株U251MG增殖的影响及可能的作用机制。[方法]用MTT比色法观察不同浓度Genistein在不同作用时间下对U251MG细胞增殖的影响,并通过流式细胞仪和免疫细胞化学染色检测p53(突变型)、c-erbB1、c-myc、cyclinD1蛋白表达。[结果]当Genistein在达到一定浓度或(和)一定时间后,对U251MG细胞增殖均有不同程度的抑制作用,且呈时间剂量依赖性。U251MG细胞经Genistein作用48h后,p53(突变型)、c-myc基因蛋白表达均较对照组明显下降(F=25.766,P﹤0.01;F=22.974,P﹤0.01),且呈剂量依赖性,而c-erbB1、cyclinD1基因蛋白表达无明显变化(F=0.984,P﹥0.05;F=2.416,P﹥0.05)。[结论]Genistein对U251MG细胞的增殖有明显的抑制作用,其抗肿瘤的机制可能与p53(突变型)和c-myc蛋白表达降低有关。
【Abstract】 [Objective]To explore the effects of genistein on proliferation of human glioma cell line U251MG and possible mechanism involved.[Methods]The effects of genistein on proliferation of U251MG cells were shown by MTT assay.The expression of p53(mutant),c-erbB1,c-myc,cyclinD1 proteins were examined with flow cytometry and immunocytochemistry.[Results]When genistein reached a certain concentration and/or a certain time,the growth of U251MG cells was inhibited in a dose-and time-dependant manner.When U251MG cells were treated with genistein for 48h,p53(mutant),c-myc proteins expression were significantly decreased compared with control group in a dose-dependent manner(F = 25.766,P﹤0.01;F = 22.974,P﹤0.01),while the c-erbB1,cyclinD1 had no obvious changes in protein expression(F = 0.984,P﹥0.05;F = 2.416,P﹥0.05).[Conclusion]Genistein can inhibit the proliferation of U251MG cells significantly,the antitumor mechanism may be related to the decreased protein expression of p53(mutant)and c-myc.
