出生前接触酒精对大鼠小脑中BDNF和TrkB的影响

【摘要】 [目的]研究出生前接触酒精对出生后大鼠小脑中脑源性神经营养因子(BDNF)及其受体酪氨酸激酶B(TrkB)的影响。[方法]选择SD母鼠24只,怀孕d6时随机法分为酒精组、正常组和代理母组。酒精组(n=6)每日摄取35卡的酒精;正常组(n=6)每日摄取与酒精组相同热卡的奶粉;代理母组(n=12)每日摄取正常的鼠食。酒精组、正常组母鼠分娩6h后,与其子分开,麻醉后心脏采血,测试血液中乙醇浓度和甲状腺素量,并作统计学分析。酒精组、正常组新生子鼠由代理母组的代理母鼠养育,并分别于生后0,7,14,21,28d(P0、P7、P14、P21、P28)时麻醉处死,采用免疫组织化学染色法,在小脑颗粒层中观察BDNF及TrkB阳性浦肯野细胞(PurkinjeCell,Pc)的分布及形态。[结果]①酒精组母鼠血液中酒精浓度明显高于正常组(P﹤0.05),而酒精组母鼠血液中甲状腺素含量明显低于正常组(P﹤0.05)。②正常组子鼠小脑中P14开始出现基本单层排列的成熟的BDNF及TrkB阳性浦肯野细胞,但酒精组子鼠小脑中始终未出现完全单层排列的成熟的BDNF及TrkB阳性浦肯野细胞,且P28时出现BDNF及TrkB阳性浦肯野细胞的明显减少。[结论]母鼠孕期摄取酒精时,引起其血液中甲状腺素含量减少,并可抑制其后代小脑中BDNF及TrkB阳性浦肯野细胞的发育。

【Abstract】 [Objective]To investigate the changes of BDNF-and TrkB in the rat cerebellar cortex with prenatal alcohol exposure.[Methods]Twenty-four time-pregnant SD rats were divided into three groups on the 6th day of gestation.Alcohol group(n = 6)received 35 calories of liquid alcohol diet every day;Control group(n = 6)was fed a liquid diet in which milk replaced alcohol isocalorically;Surrogate group(n = 12)intook food normally.Blood samples were obtained by cardiac puncture,under anesthesia,from alcohol and control groups within 6 hours after its last pup was born.The plasma concentrations of alcohol and thyroxine were determined at the Department of Clinical Laboratory,and were compared using the Kruskal-Wallis test.Pups of alcohol and control groups were fostered by surrogate mothers and executed at anesthesia at postnatal 0,7,14,21 and 28 days for measuring the distribution and form of BDNF-and TrkB-immunoreactive Purkinje cells via immunohistochemistry in the cerebellar cortex.[Results]①The blood alcohol concentration was higher in alcohol group dams than in control group,with significant difference(P﹤0.05);Thyroxine concentration was significantly decrease in alcohol group dams than in control group(P﹤0.05).②A similar developmental pattern of BDNF-and TrkB-immunoreactive Purkinje cell was observed on and after P14 in control pair-fed.These Purkinje cells of alcohol-fed group showed immature features.Single-layer arrangement of these Purkinje cells in alcohol-fed group was not completely achieved throughout postnatal life.In addition,the number of these Purkinje cell decreased at P28 as compared to control pair-fed group.[Conclusion]When dams exposure with alcohol within gestation,not only the blood thyroxine concentration was significantly decreased in dams,but also BDNF-and TrkB-synthesis of cerebellar cortex and growth of BDNF-and TrkB-immunoreactive Purkinje cells were impaired in offsprings.
