应用间期FISH研究在肾癌石蜡包埋组织中抽提细胞核的染色体异常

【摘要】 [目的]应用间期FISH研究从石蜡包埋肾癌组织中抽提细胞核的染色体异常,探讨此方法的可行性。[方法]选择20例不同类型的肾癌(8例乳头状肾癌,4例Xp11.2染色体异位性肾癌,6例肾透明细胞癌,2例肾嗜酸细胞腺瘤)的石蜡包埋组织,抽提间期细胞核,制备单细胞悬液,用CEP17探针检测17号染色体的倍体,验证此方法的有效性。[结果]在全部20例肾癌样本中成功进行了FISH检测,其中,8例乳头状肾癌存在17号染色体的三体,2例肾嗜酸性腺瘤与6例肾透明细胞癌中17号染色体倍体正常,4例Xp11.2易位性肾癌存在17号染色体的3倍与4倍体。[结论]在石蜡包埋组织中抽提间期细胞核,应用FISH检测染色体异常的方法敏感、特异、可行。

【Abstract】 [Objective]To use interphase fluorescence in situ hybridization(FISH)to study renal cell carcinoma(RCC)in extract isolated nuclei from paraffin-embedded tissue and investigate its efficacy.[Methods]The efficacy of this method was investigated in 8 papillary renal cell carcinoma(PRCC),4 Xp11.2 translocations RCC(Xp11.2RCC),6 clear cell carcinoma(CCRCC),and 2 renal oncocytoma.Using interphase FISH with CEP17 probe to study chromosomal 17 ploid in extract isolated nuclei from formalin fixed and paraffin embedded tissue.[Results]FISH studies were successful for each of the 20 specimens.Triploidy in chromosome 17 were detected in all specimens of PRCC,dipoid in chromosome 17 were detected in 8 specimens of oncocytoma,and polyploid in chromosome 17 were detected in 4 specimens of Xp11.2RCC.[Conclusion]The FISH technique is useful to detect chromosome anomalies with high sensitivity and specificity in extracted nuclei from paraffin-embedded tissue.
