EDTA提高Microtox检测有机化合物毒性灵敏度的研究

【摘要】 [目的]改善明亮发光杆菌细胞通透性、提高Microtox对有机污染物生物毒性检测的灵敏度。[方法]以乙二胺四乙酸二钠(EDTA-Na2)处理明亮发光杆菌,用疏水荧光探针N-苯基-1-萘胺(NPN)检测明亮发光杆菌通透性的改变,应用发光细菌急性毒性检测技术(Microtox)检测有机化合物在通透性改变前后的50%发光抑制浓度(EC50)。[结果]加入EDTA后菌体对NPN的吸收系数明显增加,40mM的ED-TA可使明亮发光杆菌对NPN的吸收系数增加47.7%。未经EDTA处理时,1,10-二氮杂菲、1,4-对位苯醌、双酚A、4-异丙基苯酚经Microtox检测的EC50值分别为181.2、0.071、37.6、0.79mg/L,处理后分别降为115.1、0.054、26.24、0.603mg/L。[结论]EDTA可以提高明亮发光杆菌对有机化合物的通透性,使Microtox检测生物毒性的灵敏度提高。

【Abstract】 [Objective]To improve the permeability of Photobacterium phosphoreum and the sensitivity of Microtox to toxic organic compounds. [Methods]Photobacterium phosphoreum was treatd with EDTA, permeability of Photobacterium phosphoreum was detected by NPN and 50%luminescence inhibition (EC50) of organic compounds on Photobacterium phosphoreum was determinned by Microtox before and after the change of permeability. [Results]Absorption coefficient was improved significantly with addition of EDTA and 40mM EDTA produced 47.7% increase in NPN uptake by Photobacterium phosphoreum . The EC50 values of 1,10-phenanthroline, 1,4-Benzoquinone, bisphenol A and 4-Isopropylphenol were 181.2, 0.071, 37.6 and 0.79 mg/L respectively without EDTA treatment, while EC50 values decreased to be 115.1, 0.054, 26.24 and 0.603 mg/L correspondingly with the treatment of EDTA. [Conclusion]EDTA can improve the permeability of Photobacterium phosphoreum , and the sensitivity of Microtox test for biological toxicity.
