G-CSF结合行为训练对缺血性脑损伤大鼠学习记忆及NGF表达的影响

【摘要】 目的:观察粒细胞集落刺激因子(G-CSF)结合行为训练对缺血性脑损伤大鼠学习记忆及神经生长因子(NGF)表达的影响。方法:随机选用2月龄清洁级wista大鼠24只设为假手术组A组,其余采用改良线栓法制作大鼠大脑中动脉闭塞(MCAO)2h/再灌注24小时模型,获成功模型72只随机分为3组,B组为非干预组,C组为G-CSF干预组(皮下注射G-CSF),D组为G-CSF+行为训练组(皮下注射G-CSF,同时进行水迷宫行为训练),每组24只。假手术组(A组)不插入线栓,其余操作与缺血再灌注组相同。各组分别于造模成功后1周、2周、4周、8周随机选取6只利用穿梭实验视频分析系统进行学习记忆能力评估,随后处死、病理切片,做HE染色和免疫组织化学实验,观察造模后大鼠海马区病理变化及NGF表达水平的改变。结果:造模后1周:B、C、D3组NGF表达均明显高于A组,C、D组高于B组(P<0.05),学习记忆能力均明显低于A组(P<0.01),3组之间无显著性差异(P>0.05);造模后2、4周:B组NGF表达降至基础水平,C、D组NGF表达水平及学习记忆能力均高于B组,其中D组差异性最为明显(P<0.01);造模后8周,D组NGF表达及学习记忆能力均明显高于B、C2组(P<0.01),C组NGF表达降至基础水平。结论:G-CSF结合行为训练较单一应用G-CSF不但神经保护作用明显增强,而且具有长时程保护作用,显著提高远期学习记忆能力。

【Abstract】 Objective:To investigate The effects of G-CSF combined with behavior training on learning-memory and expression of NGF after ischemic brain injury in rats.Methods:Clean conventional wista rats,aged 2 months, were selected. Twenty-four of the rats were randomly selected as sham group(group A). And the rests used A modified middle cerebral artery occlusion (MCAO) to establish a 2 h/reperfusion 24 h CIRI model in rats.Seventy-two successful MCAO were divided randomly to 3 groups.Group B was the control group of MCAO .Group C was G-CSF group.Group D was G-CSF + behavior training. The number of the rats in each group was 24. six rats were selected in each group on 1st week、2nd week、4th week and 8th week after successfully modeled, respectively.The abilities of learning and memory were detected by the shuttle experiment video analysis system.The pathological changes of the hippocampus and the expression of NGF were observed by HE and immunohistochemistry staining, respectively.Results:1st week:The expression of NGF in group B、C、D was much higher than that in group A,and group C、D>B (P<0.05),with the abilities of learning and memory much lower than group A(P<0.01).There was no significant difference among group B、C、D(P>0.05). 2nd and 4th week:the expression of NGF in group B decreased to the normal level. The expression of NGF and the abilities of learning and memory of group C、D were higher than group B (P<0.05; P<0.01). 8th week:the expression of NGF and the abilities of learning and memory of group D were much higher than group B、C(P<0.01). The expression of NGF in group C decreased to the normal level.Conclusions:The effects of G-CSF combined with behavior training on neuroprotection might be not olny strengthened , but also they be long term protective. The abilities of learning and memory in long term have been improved significantly.
