多粘芽孢杆菌Dw-6菌株培养条件的优化及其发酵液抑菌活性的初步研究
Optimization of Ferment Conditions and Antibacterial Activity Study of Paenibacillus polymyxa Dw-6
【摘要】 10倍稀释法筛选出最佳产抑菌活性物质的多粘类芽孢杆菌Dw-6菌株,试验表明Dw-6菌株最佳培养基为发酵培养基。利用4因素均匀设计方案进行培养基配方的优选,同时进行培养时间和发酵条件的优化。结果表明,Dw-6菌株优化后的培养基配方为(50 mL)蛋白胨3.83 g、酵母浸出粉3.9 g、MgSO4.7H2O0.105 g、K2HPO4.3H2O 0.17 g,最佳培养时间48 h,初始pH 7.0,培养温度30℃。经培养基优化后,Dw-6菌株在培养基初始pH 7.0时,30℃培养48 h的发酵上清液对大肠杆菌、金黄色葡萄球菌及出芽短梗霉菌均表现出很强的抑制作用。
【Abstract】 Paenibacillus polymyxa Dw-6 strain was selected by 10 fold diluted,and fermentation medium is the best culture medium to Dw-6 to secrete antagonists.Ferment conditions were optimized as follow: bran 3.83 g,powder 3.9 g,MgSO4·7H2O 0.105 g and K2HPO4·3H2O 0.17 g dissolve in 50 mL water,and the optimum time is 48h,culture medium original pH 7.0,at 30 ℃.The fermentation broth of Dw-6 has strong activity against E.coli,Staph.aureus and Aureobasidium pullulans after cultured 48 h at 30 ℃ with fermentation medium original pH 7.0.
