转反义trxs基因小麦籽粒发育过程中基因表达差异

【摘要】 为探讨反义trxs基因在转基因抗穗发芽小麦中的作用机制,以转反义trxs基因和非转基因小麦开花后5~30 d的籽粒为材料,采用mRNA差异显示技术分析了转基因与非转基因小麦在籽粒形成过程中的基因表达差异。结果表明,在籽粒发育过程中,转基因与非转基因小麦存在明显的基因表达差异,差异可归纳为转基因增强型、转基因减弱型、转基因特异型和沉默型4种类型,而且特异型表达明显高于增强型和减弱型;差异条带数随种子成熟度的增加而不断增多,到花后第25天达到最高(77条),然后略有下降,且在胚乳中的差异明显高于胚中的。

【Abstract】 In order to clarify the function mechanism of antisense-thioredoxins(anti-trxs) gene in transgenic wheat with pre-harvest sprouting resistance characteristics,the gene expression patterns in transgenic line 01TY70-1-17-5 and non-transgenic cultivar ’Yumai 70’ were detected using 5-30 d seeds after pollination by mRNA differential display during seed filling stage.The results showed that there were significant differences in gene expression between transgenic line and non-transgenic controls during seed development.The differences could be summarized into four types,namely,enhanced expression,decreased expression,specific expression and silence expression.Moreover,the specific expression type was significantly higher than the enhanced and reduced expression type.The specific bands increased with seed maturity and reached the maximum(77) 25 days after pollination,and then decreased slightly;The difference in the endosperm was higher than in the embryos.
