【摘要】 目的:探讨芹菜素对人前列腺癌PC-3细胞裸鼠移植瘤凋亡相关蛋白Bax、Bcl-2表达的影响。方法:通过皮下种植PC-3细胞建立人前列腺癌裸鼠移植瘤模型,成瘤后随机分为4组:对照组(DMSO+NS,0.2 mL/d)、芹菜素低剂量组(12.5 mg.kg-1.d-1)、芹菜素中剂量组(25 mg.kg-1.d-1)、芹菜素高剂量组(50 mg.kg-1.d-1),每组6只,每日腹腔注射1次,共28次。应用免疫组织化学法检测裸鼠移植瘤组织凋亡相关蛋白Bax、Bcl-2的表达变化。结果:免疫组织化学法结果显示,芹菜素中、高剂量组能促进Bax蛋白的表达,抑制Bcl-2蛋白的表达,两组与对照组和低剂量组相比差异有统计学意义(P<0.05)。但芹菜素低剂量组Bax和Bcl-2蛋白的表达和对照组相比差异无统计学意义(P>0.05)。结论:中、高剂量组芹菜素能上调前列腺癌裸鼠移植瘤Bax蛋白的表达,下调Bcl-2蛋白的表达。更多还原

【Abstract】 Objective: To study the effect of apigenin on apoptosis-related protein Bax and Bcl-2 expression of human prostate cancer PC-3 cells transplanted tumor in nude mice.Methods: Human prostate cancer PC-3 cells cultured in vitro were subcutaneously inoculated into nude mice to establish transplanted tumor model.All tumor-bearing mice were randomly divided into 4 groups:control group(DMSO and normal saline,0.2 mL/d),low dose apigenin group(12.5 mg·kg-1·d-1),moderate dose apigenin group(25 mg·kg-1·d-1) and high dose apigenin group(50 mg·kg-1·d-1),there was 6 nude mice in each group,daily intraperitoneal injected,a total of 28 times.The expression of Bax、Bcl-2 in tumor tissue was determined with immuno-histochemical methods.Results: The expression of Bax in moderate and high group were higher than that of control group and low dose group(P <0.05),while the expressions of Bcl-2 in moderate and high group were lower than that of control group and low dose group(P <0.05).There was no such change seen between control and low dose group(P >0.05).Conclusion: The moderate and high dose of apigenin can up-regulate the expression of Bax protein,and down-rugulate the expression of Bcl-2 protein on the growth of transplanted tumor of prostate cancer PC-3 cell in nude mice.更多还原
