丙烯酰胺染毒小鼠海马组织结构和Caspase-3、GFAP表达的变化

【摘要】 目的探讨丙烯酰胺染毒对雄性小鼠海马组织结构和Caspase-3、GFAP表达的影响。方法健康雄性昆明种小鼠40只,随机分为正常对照组和低、中、高浓度3个剂量实验组,每组10只。对照组小鼠腹腔注射生理盐水;实验组小鼠分别代以5、30和60 mg/(kg.d)丙烯酰胺水溶液腹腔注射。在10d时分别处死各组小鼠,快速取出海马,Zamboni固定液固定,石蜡包埋,常规制片,苏木素-伊红(HE)染色,Caspase-3和GFAP免疫组织化学染色,光学显微镜观察。结果实验组小鼠海马同对照组相比显微结构未见明显改变。与对照组比较,各剂量组小鼠海马神经元Caspase-3阳性表达明显增强(P<0.01),但各剂量组间未见明显改变(P>0.05);各剂量组小鼠海马胶质细胞GFAP阳性表达明显增强(P<0.01),高剂量组表达较低、中剂量组增强(P<0.05)。结论丙烯酰胺染毒可造成小鼠海马神经元和神经胶质细胞损伤。

【Abstract】 Objective To study the changes on the microstructure and the expressions of Caspase-3 and GFAP in mouse hippocamus after acrylamide exposure.Methods Fourty healthy male mice of Kunming Genus were divided into four groups at random: control group(n=10),5,30,60 mg/(kg·d) treatment groups(n=10).Physiological saline was administrated intraperitoneally(ip) to mice of control group.while acrylamide solution was administrated intraperitoneally(ip) to mice of treatment groups.All mice were euthanized on the 10th day and hippocampus was dissected out to be fixed in the Zamboni fixation solution,embed in Paraffin,produced the section,and stained with HE or by immunohistochemistry stain.Results Compared with the control group there were no significant changes on microstructure in hippocampus.But there was an significant increase of the Caspase-3 and the GFAP expression of the hippocampal neurons in each treatment group.There are not significant change in each dose groups.Compared with the control group,the GFAP expression of the hippocampal Glial cell significantly increase in each dose groups,the GFAP expression of high dose group is stronger than the expression of low and middle dose group.Conclusion Acrylamide could damage the neurons and glial cells in mouse hippocamus.
