二乙氧基乙醇雄性大鼠生殖系统毒性研究

【摘要】 目的观察研究二乙氧基乙醇(2-Ethoxyethanol,2-EE)染毒雄性大鼠睾丸和附睾毒性、精子毒性、以及性行为与妊娠率的改变,探讨各生殖毒性指标之间的关联性,为确定2-EE雄性生殖毒性健康监护指标和诊断指标,开展2-EE职业健康危险度评价提供科学依据。方法选择清洁级雄性Wistar大鼠,分别给予剂量200、400和800 mg/kg的2-EE,连续灌胃13 d,观察其睾丸和附睾脏器系数、病理改变;精子计数和精子活力改变;染毒雄鼠与正常雌鼠合笼,观察其性行为和配对雌鼠妊娠率的改变。对照组以等量生理盐水灌胃。结果 800 mg/kg染毒组睾丸脏器系数明显下降(P<0.01),各级生精细胞明显减少,精子活力下降(P<0.05);400 mg/kg以上染毒组雄鼠骑跨潜伏期、插入潜伏期、射精潜伏期和射精后间隔期不同程度延长,差异无显著性统计学意义(P>0.05);雌鼠妊娠率与对照组比较差异无显著性统计学意义(P>0.05)。结论 2-EE染毒雄鼠睾丸毒性明显,精子毒性表现滞后于性行为改变且毒效应剂量较高,性行为改变可作为2-EE雄性生殖毒性直观而敏感的效应指标。

【Abstract】 Objective To observe and research the testis and epididymis toxicity,sperm toxicity,and changes in sexual behavior and pregnancy rate,discuss the relationships between them in the male rats exposed to 2-Ethoxyethanol(2-EE) for exploring sensitive occupational health surveillance and diagnosis indicatiors and providing scientific basis for the further occupational health risk assessment.Method The experimental groups were treated with 200,400,800 mg/kg 2-EE by gavage for 13 days,the negative control group was treated by 0.9% saline.To observe the signs of poisoning,organ coefficient and pathology of testis and epididymis,sprem count and activity,sexual behavior and female pregnancy rate.Results In comparison with negative control group,organ coefficient of testis(P < 0.01) and sperm activity(P < 0.05) declined significantly in 800 mg/kg group.The mount latency,intromission latency,ejaculation latency and postejaculatory interval were extended in 400 mg/kg and 800 mg/kg groups,but there were no significant differences(P > 0.05).Conclusion The testis toxicity is significant,the sperm toxicity lags behind the changes in sexual behavior and the effect dose is higher.The change in sexual behavior is the directly sensitive indicator to male reproductive toxicity by 2-EE.
