卵白蛋白致敏时减毒活菌卡介苗干预对哮喘小鼠气道炎性反应的影响

【摘要】 目的探讨分别在卵白蛋白致敏时和致敏前以不同剂量接种卡介苗对哮喘小鼠气道炎性反应及黏液分泌的影响。方法将C57BL/6小鼠随机分为模型组、阴性对照组、4组卡介苗接种组。10~5、10~6集落形成单位致敏前接种依次为A1组、A2组,致敏时接种依次为B1组、B2组。收集肺泡灌洗液和外周血,分别测定其中IL-5、IFN-γ浓度;病理观察肺组织炎性反应及气道黏液分泌情况,测算杯状细胞化生指数、上皮黏液储备指数。结果卡介苗接种后,只有B1组、B2组能抑制化生指数(P<0.05);B2组较A1组、A2组明显地降低了储备指数(P<0.05),降低了肺泡灌洗液中IL-5/IFN-γ比值(P<0.01)。结论致敏时接种卡介苗较致敏前接种能更有效抑制哮喘小鼠气道炎性反应、减少黏液分泌,并且存在一定量效关系,可能与纠正Th1/Th2失衡有关。

【Abstract】 Objective To investigate the effect of intraperitoneal BCG on the airway inflammation and mucus production of a mouse model of asthma vaccinated with BCG twice before and during allergen sensitization at different doses.Methods Male C57BL/ 6 mice were sensitized twice and challenged three times by ovalbumin as asthma model group,and mice were treated by normal saline as control group.BCG was intraperitoneally vaccinated twice before and during allergen sensitization with 105,and 106 CFUs.Bronchoalveolar lavage fluid(BALF) and peripheral blood(PB) were collected.IL- 5,and IFN-γin BALF and PB were measured by ELISA.The histology of lung tissues was also observed.Results Goblet cell hyperplasia ratios were restrained only in Bl and B2 groups compared with model group(P<0.05 ).The mucus occupying ratio(MOR) of epithelial cells in B2 group decreased more significantly than in A1 and A2 groups(P<0.05).For IL- 5/ IFN-γin BALF,significant difference was found between B2 group and A1,A2 groups(P<0.01 ).Conclusions Intraperitonally vaccinated BCG during sensitization inhibits the mucus production in the airway epithelium more significantly than before sensitization,and in a dose- dependent manner,which may be related to Th1/Th2 balance in the asthma model adjusted by BCG vaccination.
