【摘要】 目的建立一种简便、快速、灵敏的反相高效液相色谱(HPLC)紫外检测法测定血清氯氮平药物浓度。方法选用C18柱,波长254nm,移动相为甲醇∶乙腈∶水∶正丁胺∶冰乙酸(16∶36.5∶47.5∶0.5∶1,调PH8.0),流速1.0ml/min,以乙醚为萃取液。结果氯氮平保留时间为(3.74±0.02)min,日内和日间平均回收率为99.2%～99.8%和99.5%～100.6%。CV分别<4.3%和5.1%,最低有效检测浓度为20ng/ml。结论本法操作简便、快速而精确,具有一定的实用价值。更多还原

【Abstract】 Objective To develop a simple and rapid HPLC method for the determination of the blood level of clozapine.Methods The analysis involved a C18 column as stationary phase and methanol,acetonitrile,water,n-Butylamine,acetic-acid(16∶36.5∶47.5∶0.5∶1)as mobile phase.The flow rate was 1.0ml/min,UV detection wavelength was 254nm.The serum samples were extracted with ethy1 ether.Results Intra-day and inter-day CV were less than 4.3% and 5.1% respectively.The recovery rates of intra-day and inter-day were 99.2%~99.8% and 99.5%~100.6% respectively.Quantification limites 20ng/ml.Conclusion The method is simple,rapid,sensitive and accurate for clinical practice.更多还原
