荧光活性染料DiO在观察成骨诱导脂肪干细胞在脱钙骨材料上的生长与分布中的应用

【摘要】 目的探讨荧光活性染料DiO对人脂肪干细胞(hADSCs)染色标记的效果及标记后的hADSCs在部分脱钙骨(PDBM)上的生长粘附情况。方法利用扫描电镜观察了细胞材料复合后细胞的生长情况;检测了DiO标记的hADSCs和DiO未标记的hADSCs在PDBM上粘附率,采用激光共聚焦显微镜观察观察了DiO标记的hADSCs在平面培养及在PDBM上的生长、粘附情况。结果 DiO标记的hADSCs平面培养时可以良好地生长,DiO标记的hADSCs粘附率为(94.5±2.2)%,未标记的hADSCs粘附率为(95.1±1.3)%,统计学分析两者无显著差异(P>0.05)。DiO标记的hADSCs在PDBM支架材料上可以良好的粘附生长。结论 DiO对被标记细胞的存活、生长无影响、不影响细胞粘附,能反映细胞在支架材料上的正常生长状况。DiO标记是一种简洁﹑准确的观察hADSCs细胞在支架材料上粘附及生长状况的方法。

【Abstract】 Objective: To investigate the effect of Dio on labeled hADSCs and its growth on partially demineralized bone matrix(pDBM).Methods: The growth of Dio-labeled hADSCs on pDBM was observed with Scanning Electron Microscope(SEM) and Laser Scanning Confocal Microscope(LSCM) respectively,and then the adhesion rate of Dio-labeled hADSCs and unlabeled ones were tested.Results: Dio-labeled cells grew fine in vitro,and there was no significant difference between the Dio-labeled cells and unlabeled ones in adhesion rate with(94.5±2.2)% and(95.1±1.3)% respectively(P>0.05).Moreover,Dio-labeled cells grew well on the surface of pDBM.Conclusion: Dio can reflect the normal state of labeled cells with no influence on the survival,growth and adhesion rate of labeled cells.Thus,Dio is a kind of easy and precise method to observe the adhesion and growth state of hADSCs on scaffolds.
