CD151基因对前列腺癌细胞增殖、转移的促进作用及其机制
CD151 Gene Delivery Promotes Proliferation and Migration of Prostatic Carcinoma Cells
【摘要】 目的研究人前列腺癌细胞株PC3细胞转染CD151基因后其增殖和迁移能力的变化及机制。方法用FuGENE HD分别将质粒pAAV-CD151(CD151组)、pAAV-GFP(GFP组)转染入PC3细胞,并设加入等体积PBS的对照组。48 h后采用Western blot检测CD151蛋白、磷酸化细胞外信号调节激酶(ERK)和总ERK的表达。采用MTT法检测CD151基因对PC3细胞增殖的影响,采用Boyden趋化小室研究CD151基因对PC3细胞迁移的影响。结果与对照组和GFP组相比,CD151组的CD151蛋白表达水平明显增加(均P<0.05);磷酸化ERK的表达量亦明显高于其它两组;细胞增殖能力比对照组和GFP组明显增高[相对吸光度值分别为:(0.245 7±0.006 4)vs(0.175 2±0.007 4)、(0.179 0±0.008 4)];细胞迁移数(107.8±9.6)亦显著高于对照组和GFP组[(53.0±7.6)和(57.0±5.2),P<0.05]。结论 CD151在前列腺癌细胞的增殖、转移中起着重要作用,是肿瘤转移的重要分子基础,其分子机制可能是CD151对ERK信号通路的激活。
【Abstract】 Objective To determine the effects of CD151 on the proliferation and migration of prostatic carcinoma cell line PC3 cells and the mechanism involved.Methods pAAV-CD151 and pAAV-GFP were constructed and transiently transfected into PC3 cells mediated with FuGENE HD.After transfection,the expression of CD151 and extracellular signal-regulated kinase(ERK)was detected by using Western blot.Cell proliferation was measured by using MTT method and cell migration by using Boyden chambers,respectively.The signaling mechanisms were assessed by using Western blot.Results Transfection of pAAV-CD151 could increase the expression of CD151 in PC3 cells.MTT method revealed that the absorbance(A)values in CD151 group(0.245 7±0.006 4)was significantly higher than in control group(0.175 2±0.007 4)and GFP group(0.179 0±0.008 4)(P<0.05).The number of migrating cells was significantly increased in CD151 group(107.8±9.6),as compared with control group(53.0±7.6)and GFP group(57.0±5.2)(P<0.05).In addition,CD151 transfection could increase the phosphorylation of ERK.Conclusion CD151 gene delivery promotes the proliferation and migration of PC3 cells.The molecular mechanism involving may be that over-expression of CD151 up-regulated the phosphorylation of ERK.
