【摘要】 目的探讨晚期扩张型心肌病(DCM)患者心肌连接蛋白43(Cx43)基因表达水平及其意义。方法 DCM病例及标本是2002～2003年华中科技大学同济医学院附属同济医院对4例晚期DCM患者进行心脏移植手术切除心脏的心肌;其中男2例、女2例,年龄分别为14、44、45和55岁。对照组4例为同期颅脑损伤和毒品中毒致死者,均为男性,年龄分别为24、29、30和37岁;死因为颅脑损伤3例、毒品中毒1例。每例心脏进行常规病理学检查并取材后,应用逆转录PCR技术定量检测左心室心肌Cx43基因表达水平,并进行比较分析。结果 DCM患者心肌Cx43基因定量检测结果为(0.188±0.167),对照组为(0.440±0.096),两组比较差异有统计学意义(P=0.025)。结论晚期DCM患者心肌Cx43基因表达水平下降,这种心肌Cx43基因表达水平下降可能与DCM患者发生心衰、心律失常和猝死有关。更多还原

【Abstract】 Objective To investigate the significance and expression level of connexin43(Cx43) gene in myocardia of patients with dilated cardiomyopathy(DCM).Methods The myocardial samples of 4 cases were obtained in the left ventricular walls of hearts transplanted from the DCM patients who hospitalized in Tongji Hospital from December 2002 to July,2003.There were 2 males and 2 females with the age being 14,44,45 and 55 years old respectively.The myocardial samples of 4 male cases in control group were obtained from the cases died of craniocerebral injury(3 cases) and poison poisoning(one case) during the same period with the age ranging 24-37 years old.Every heart was examined with routine pathological method.The expression of Cx43 gene in left ventricular myocardia of both groups was detected quantitatively by using RT-PCR,and all data of two groups were statistically analyzed.Results The expression of Cx43 gene in patients with DCM and control group was(0.188±0.167) and(0.440±0.096) respectively,with significant difference(P=0.025).Conclusion The expression of Cx43 gene in myocardia of DCM patients is declined,which may contribute to heart failure,arrhythmia and sudden death of patients with DCM.更多还原
