【摘要】 目的探讨枸杞多糖(LBP)诱导人肺癌A549细胞的凋亡的作用及其作用机制。方法在体外培养的肺癌A549细胞株中加入不同浓度的LBP,应用流式细胞仪FITC/PI双染法检测细胞凋亡率,RT-PCR及Western blot法分析加药前后凋亡抑制基因Survivin mRNA和蛋白表达情况。结果流式细胞仪FITC/PI双染法检测结果显示LBP可诱导人肺癌A549细胞凋亡,并且随着LBP浓度增加,凋亡率逐渐增加;LBP可使Survivin mRNA和蛋白的表达明显降低,并成一定的量效关系。结论 LBP可显著诱导人肺癌A549细胞凋亡,其作用机制可能与LBP抑制A549细胞凋亡抑制基因Survivin表达有关。更多还原

【Abstract】 Objective To investigate the effect and mechanism of LBP on apotosis of human lung adenocarcinoma cells A549. Methods After A549 were treated with LBP in different concentrations and with different time,the Annexin V-FITC/PI double staining analysis by flow cytometry was used to evaluate apoptosis rate.The levels of Survivin mRNA and Survivin protein were detected by RT-PCR and Western Blot. Results LBP could induce cell apoptosis in A549 cells,the apoptotic rate was dose depedent.The survivin gene and protein expression was down regulated with LBP,which was LBP dose depedent. Conclusion LBP can effectively induce cells apoptosis in lung adenocarcinoma A549 cells,this may result from its actioin of reduce survivin gene expression.更多还原
