杞荷制剂防治非酒精性脂肪性肝炎的实验研究
Experimental research on the Preventive Effect of Qihe Preparations against Nonalcoholic Steatohepatitis
【摘要】 目的探讨杞荷制剂防治非酒精性脂肪性肝炎(NASH)的作用及其机制。方法采用高脂饮食复制大鼠NASH模型。Wistar大鼠72只随机分为正常对照组、空白模型组、杞荷制剂小、中、大剂量组、复方蛋氨酸胆碱片组,每组12只。观察动物一般情况,计算肝指数、肝组织细胞脂肪变性、炎症活动程度,测定血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、血清甘油三酯(TG)、总胆固醇(TCH)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、肿瘤坏死因子-α(TNF-α),肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)等的变化。结果与空白模型组相比,杞荷制剂小、中、大剂量组大鼠肝指数明显降低,炎症活动程度皆有明显改善;血清ALT、AST、TG、TCH、LDL明显降低(P<0.01或P<0.05);肝中SOD活性显著升高(P<0.01)、MDA含量显著降低(P<0.01);血清TNF-α的含量显著减少(P<0.01),明显优于复方蛋氨酸胆碱片对照组(P<0.01或P<0.05)。结论杞荷制剂可能通过改善肝功能、调节脂肪代谢以及缓解氧化应激、抑制致炎细胞因子的表达,起到防治NASH的作用。
【Abstract】 Objective To explore the function and mechanism of Qihe preparations to prevent nonalcoholic steatohepatitis(NASH). Methods Using NASH model of high fat diet rats.72 Wistar rats were only randomly divided into normal control group,blank model group,Qihe preparations of small,medium and large dose groups,compound methionine choline piece group,and each group with 12.Observe the general conditions of the animals,calculate the liver index,liver tissue cells steatosis,the activity level of inflammation,and determine serum alanine aminotransferase(ALT),aspartic acid aminotransferase(AST),serum triglyceride(TG),total cholesterol(TCH),high-density lipoprotein(HDL),and low-density lipoprotein(LDL),tumor necrosis factor alpha(TNF-α),liver SOD,malondialdehyde(MDA) and other elements’ changes. Results Compared with blank model group,the liver index of Qihe preparations of small,medium and large dose groups of rats decreased significantly,and the activity degree of inflammation was obviously improved;Serum ALT,AST,TG,TCH,LDL reduced significantly(P<0.01 or P<0.05);Liver SOD activity increased(P<0.01),the content of MDA was significantly reduced(P<0.01);the content of Serum TNF-α was significantly reduced(P< 0.01),obviously better than the compound methionine choline piece in control group(P< 0.01 or P<0.05). Conclusion Qihe preparations may play the role of the prevention and control of NASH through improving liver function,adjusting fat metabolism,alleviating oxidatie stress,and inhibiting the expression of inflammatory cells cytokines.
