海洋药用红藻龙须菜高效液相色谱指纹图谱的研究

【摘要】 目的建立海洋药用红藻龙须菜高效液相色谱(HPLC)指纹图谱,为其真伪鉴别和质量控制提供科学依据。方法采用反相Extend C18色谱柱,以乙腈-0.1%(V/V)磷酸水为流动相,二元线性梯度洗脱,流速为1.0 ml/min,柱温为20℃,检测波长为226 nm,进样量为30μl;采用高效液相色谱-质谱联用技术(HPLC-MS)对其特征峰进行快速分析;通过相似度分析对8批龙须菜进行评价。结果以8批龙须菜样品的6个共有峰为评价指标,建立了龙须菜HPLC指纹图谱,相似度均在0.90以上;通过HPLC-MS分析获得4个特征峰的质谱信息,实现了龙须菜指纹图谱共有峰和特征峰的准确定位。结论该方法简单可靠、稳定性好,是龙须菜真伪鉴别和质量控制的有效方法。

【Abstract】 ObjectiveTo establish a high performance liquid chromatography(HPLC) fingerprint method for the quality evaluation and discrimination of oceanic officinal red alga-Gracilaria lemaneiformis(Rhodophyta). MethodsThe chromatographic fingerprint of the 8 samples was determined by injecting 30 μl of the sample solution each time on a reversed phase extend-C18 column with the gradient elution solvent system composed of 0.1% phosphatic solution and acetonitrile.The flow rate was 1.0 ml/min,the column temperature was maintained at 20 ℃ and the detection wavelength was at 226 nm.Characteristic peaks of the fingerprint could be rapidly analyzed by high performance liquid chromatography-mass spectrometry(HPLC-MS),and the method of similarity analysis was used to evaluate the 8 samples. ResultsThe HPLC fingerprint of Gracilaria lemaneiformis(Rhodophyta) was set up,which included 6 common peaks,and the similarities of 8 samples were all better than 0.90.Mass spectra of the 4 characteristic peaks obtained by HPLC-MS could be used to accurately confirm the characteristic and common peaks in fingerprint of Gracilaria lemaneiformis(Rhodophyta). ConclusionThe method is simple and stable,and it can be used for identification and quality control of Gracilaria lemaneiformis(Rhodophyta).
