安心颗粒对慢性心衰大鼠细胞因子和心肌重构的影响

【摘要】 目的观察安心颗粒对慢性心力衰竭(CHF)模型大鼠细胞因子和心肌重构的影响。方法将50只雌性SD大鼠随机分为正常对照组(A组)、模型组(B组)、卡托普利组(C组)、安心颗粒低剂量组(D组)、安心颗粒高剂量组(E组),每组10只。B~E组采用阿霉素腹腔注射造模,每周1次,连续6周。造模同时,C~E组分别按6.25 mg.kg-1.d-1,0.8 g.kg-1.d-1,1.6 g.kg-1.d-1剂量灌胃给药。造模结束后,分别测定各组血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)水平及左室重量指数(LVMI)。结果模型组血清TNF-α、IL-6水平及LVMI均显著性升高(P<0.01);安心颗粒低、高剂量组均可显著性降低模型大鼠血清TNF-α、IL-6水平及LVMI(P<0.05或P<0.01),且呈现一定剂量效应关系。结论安心颗粒可通过调控炎性细胞因子、抑制或延缓心肌重构,防治心力衰竭。

【Abstract】 ObjectiveTo study the effect of Anxin granules on cytokines and myocardical remodeling in rats with heart faliure.MethodsFifty female SD rats were randomly divided into five groups:normal control group(A),model group(B),captopril group(C),low-dosage AXG group(D) and high-dosage AXG group(E).CHF model was established by intraperitoneal injection of low-dose of adriamycin,once a week for continuous 6 weeks.Meanwhile,group C was given captopril injection(6.25 mg·kg-1·d-1),and groups D and E received gastric gavage of low-dosage AXG(0.8 g·kg-1·d-1) and high-dosage AXG(1.6 g·kg-1·d-1) respectively.After 6 weeks of modeling,serum levels of TNF-α and IL-6 in different groups were tested by ELISA,and the left ventricular mass index(LVMI) was measured.ResultsCompared with group A,the serum levels of TNF-α and IL-6,and LVMI were markedly increased in model group(P<0.01).Low-and high-dosage AXG decreased the serum levels of TNF-α and IL-6 as well as LVMI of model rats(P<0.05 or P<0.01),in a dose-dependent manner.ConclusionAXG can alleviate the ventricular remodeling in CHF by regulating the inflammatory factors,restrain the development of CHF,and serves as a remedy for prevention and treatment of CHF.
