维胃方治疗大鼠实验性胃溃疡的尿液代谢组学研究
Metabonomic Investigation of Urine from Experimental Gastric Ulcer Rats Treated with Weiweifang Recipe
【摘要】 目的探讨维胃方治疗大鼠实验性胃溃疡的尿液代谢表型的变化及相关机制。方法采用乙酸灼烧法制备大鼠胃溃疡模型;运用气相色谱-质谱(GC-MS)获得正常对照组、模型组、自愈组与维胃方高、中、低剂量组的大鼠尿液代谢物谱。运用偏最小二乘法判别分析(PLS-DA),寻找表征正常对照组与模型组差异的生物标记物;使用主成分分析(PCA)考察维胃方不同剂量组对胃溃疡的治疗效果,并根据标记物水平的变化解释相关机制。结果模型组与正常对照组的尿液代谢物谱具有显著差异;获得的生物标记物包括有机酸、氨基酸等,两组标记物水平有显著性差异(P<0.01或P<0.05)。维胃方不同剂量组的物质水平出现不同程度回调,以中剂量组幅度最大。结论胃溃疡大鼠出现代谢异常,维胃方通过调节机体的代谢机能有效治疗胃溃疡,且以中剂量为佳;运用代谢组学技术可从整体上揭示中药对疾病的治疗作用与机制。
【Abstract】 Objective To investigate the changes of urinary metabolic phenotype and their related mechanisms in experimental gastric ulcer rats treated with Weiweifang(WWF,a kind of Chinese medicine recipe).Methods Forty-eight male Wistar rats were randomly divided into 6 groups: control group,model group,spontaneously healing group and WWF treatment groups(high-dose,middle-dose and low-dose groups),with 8 rats in each group.Gastric ulcer was caused by serosal application of acetic acid in rats and metabolic spectra of urine from each group were acquired with GC-MS technique.Then the processed data of control group and model group were subjected with least squares discriminant analysis(PLS-DA) to discover biomarkers related to gastric ulcer.Principal component analysis(PCA) including all groups was performed to assess therapeutic effect of WWF on gastric ulcer,and the mechanisms were explained according to the changes of biomarkers’ levels.Results The urinary metabolic spectra were different from each group.The biomarkers acquired included organic acids,amino acids and so on,and their levels were significantly different between normal group and model group(P<0.01 or P<0.05).While in WWF groups,they were trending toward normal in certain degree,especially middle-dose group.Conclusion The metabolisms in gastric ulcer rats were abnormal and middle dosage WWF was more effective on gastric ulcer treatment by metabolism regulation.The therapeutic effect and its mechanism of traditional Chinese medicine can be revealed in global aspect by metabonomics.
