强心益气汤抗心肌肥厚作用以及对RAAS、CaN信号通路的影响
Effects of Qiangxinyiqi Decoction on Myocardial Hypertrophy and Mechanisms Involved in RAAS,CaN Singnal Transduction in Rats
【摘要】 目的观察强心益气汤(Qiangxinyiqi Decoction,QXYQD)对压力超负荷大鼠左室肥厚的防治作用,并从肾素-血管紧张素-醛固酮系统(RAAS)和钙调神经磷酸酶(CaN)角度探讨其作用机制。方法采用腹主动脉部分结扎术制备大鼠心肌肥大模型。48只SD大鼠随机分成假手术组,模型对照组,低、高剂量强心益气汤组(24,72 g.kg-1.d-1)。术后第4周开始灌胃给药,连续4周。分别测定血浆中血管紧张素Ⅱ(Ang Ⅱ)、醛固酮(ALD)含量及CaN活性;并测定左室质量指数(LVMI)、心肌细胞直径(MD)、胶原容积分数(CVF)。结果强心益气汤呈剂量依赖性抑制模型大鼠LVMI、MD和CVF的增加,并可降低模型大鼠血浆中Ang Ⅱ、ALD的含量和CaN的活性。结论强心益气汤可对抗腹主动脉结扎所致心脏重构的作用,能抑制心肌细胞的肥大,其机制可能与减少Ang Ⅱ、ALD的含量和降低CaN活性有关。
【Abstract】 Objective To observe the effects of Qiangxinyiqi Decoction(QXYQD) on myocardium remodeling and to explore the mechanisms in rats with pressure overload.Methods Pressure overload rat model was established by partly constricting the abdominal aortas.Forty-eight adult male SD rats were randomized into 4 groups: sham operation group(SOG),operation group(OG),low-dose QXYQD group(L-QXYQD G,24 g·kg-1·d-1),high-dose QXYQD group(H-QXYQD G,72 g·kg-1·d-1).L-QXYQD G and H-QXYQD Gs were orally administered QXYQD once a day from 4th to 8th weeks.The other 2 groups were given the same amount of normal saline in the same way.The contents of angiotensin(Ang Ⅱ),aldosterone(ALD) and activity of calcineurin(CaN),and left ventricular weight index(LVMI) of the four groups were recorded;cardiac myocyte diameters(MD) and collagen volume fraction(CVF) were determined by video densitometry the 8th week.Results(1) Compared with SOG group,the Ang Ⅱ,ALD and CaN in OG group increased significantly;compared with OG group,those of treatment groups were decreased significantly.(2) Compared with SOG group,the content of collagen accompanied by the myofibrilla in OG group,the LVMI,MD and CVF were higher with significant difference;compared with OG group,those of treatment groups were lower with significant difference.Conclusion Qiangxinyiqi Decoction has beneficial effects on myocardium remodeling,and can lighten and postpone the accumulation of myocardial collagen of rats with pressure overload;the mechanism involved in decreasing the contents of Ang Ⅱ,ALD and activity of CaN.
