丹参素对过氢化氢所致人脐静脉内层细胞损伤的保护作用研究

【摘要】 目的研究丹参素(Tanshinol)对过氧化氢所致人脐静脉内皮细胞(ECV-304)损伤的保护作用。方法噻唑蓝(MTT)法检测细胞活力,比色法测定细胞培养液中超氧化物歧化酶(SOD)活性及细胞内丙二醛(MDA)含量,RT-PCR方法测定细胞内皮素1(ET-1)mRNA,过氧化物酶体增殖物激活受体γ(PPAR-γ)mRNA,血栓调节蛋白(TM)mRNA的表达,免疫细胞化学方法测定TPA,PAI-1含量。结果丹参素可剂量依赖性地抑制过氧化氢所致的细胞活力降低,减少MDA生成,提高SOD活性,并可抑制ET-1mRNA的表达,升高PPAR-γmRNA,TMmRNA的表达,增加TPA蛋白表达,减少PAI-1蛋白表达。结论丹参素对过氧化氢所致ECV-304损伤具有明显的保护作用。

【Abstract】 Objective To study the protective effects of tanshinol on human umbilical vein endothelial cell(ECV-304) injury induced by hydrogen peroxide in vitro.Methods The activity of ECV-304 was monitored by MTT assay.The activity of SOD level in culture medium was measured by colorimetric,so was MDA content of ECV-304.The expression of peroxisome proliferator-activated receptor γ,ET-1and thrombomoduline was measured by means of reverse transcription polymerase chain reaction.The expression of tissue plasminogen activator and plasminogen activator inhibitor 1 was monitored by the way of immuocytochemistry.Results Tanshinol inhibited the activity of ECV-304 injured by hydrogen peroxide,reduced MDA content and enhanced the activity of SOD.In addition it could also reduce the expression of ET-1 and plasmingen activator inhibion,increase the expression of peroxisome proliferators-activated receptor γ and thrombomoduline and tissue plasminagen activator in the ECV-304.Conclusion Tanshinol has obvious protective effects on ECV-304 injury induced by hydrogen peroxide.
