正交试验优化中药萹蓄中总黄酮的提取工艺

【摘要】 目的利用索氏提取法提取萹蓄中总黄酮,并应用分光光度法对总黄酮含量进行测定。方法以总黄酮为考察参数,通过正交试验优化提取工艺。结果体积分数为7 0%的乙醇溶液、提取时间为1.5 h、提取温度85℃、m(萹蓄)∶V(乙醇)=1 g∶40 L时为最佳提取工艺。结论通过最佳工艺条件下测定萹蓄中总黄酮的萃取率为15.40%,总黄酮含量质量分数为0.308%,回收率在99.8%~102.4%之间,RSD=0.957%(n=5)。

【Abstract】 Objective To use the Soxhlet extractor for the extract of total flavonoids from Polygonum aviculare and determine the flavonoid content in Polygonum aviculare by UV spectrophotometry.Methods The best extraction technology was obtained by the way of orthogonal experiment according to the content of total flavonoids.Results The optimum conditions were as follows:the extraction solution concentration was 1∶ 40(1 g of Polygonum aviculare:40 L of 70% ethanol),extraction solvent was 70% ethanol,extraction temperature was 85 ℃,extraction time was 1.5 h.Conclusions The best technology for the extraction of total flavonoids is obtained with extract ratio of 15.40%,the content of total flavonoids is 0.308%,the recovery is between 99.8% and 102.4%,RSD is 0.957%(n=5).
