人肺腺癌细胞系SPCA1边缘细胞亚群的初步分析

【摘要】 目的检测和分选人肺腺癌细胞系SPCA1中的边缘细胞,并对其表面标志进行初步分析。方法用流式细胞仪检测和分选SPCA1中的边缘细胞和主群细胞,并用细胞免疫化学和激光共聚焦检测ABCG2和CD133在SPCA1中的表达。结果边缘细胞亚群占SPCA1细胞的0.7%,维拉帕米阻断后减少至0.0%。细胞免疫化学结果显示ABCG2和CD133在SPCA1的边缘细胞中高表达,而在未分选的SPCA1细胞和主群细胞中则低表达。结论人肺腺癌细胞系SPCA1中存在边缘细胞,提示肺癌干细胞的存在。

【Abstract】 Objective To identify the side population(SP) cells isolated from human lung adenocarcinoma cell line SPCA1,and to investigate the marker of SP cells.Methods SP cells and main population cells were measured and sorted by FACS from SPCA1,and the expression of ABCG2 and CD133 in SPCA1 was detected with immunocytochemistry and immunofluorescence technic.Results SP cells accounted for 0.7% of the SPCA1 cells,which were decreased to 0.0% by treating with verapamil.Results obtained by the immunocytochemical method showed that ABCG2 and CD133 were highly expressed in side population cell of SPCA1,but the levels of ABCG2 and CD133 were low in the unsorted cells and the main population cells of SPCA1.Conclusion Human lung adenocarcinoma cell line SPCA1 contains SP cancer stem-like cells which can be decreased by verapamil.
