应用芯片技术对非霍奇金淋巴瘤标志物的研究

【摘要】 目的应用蛋白质组学技术寻找非霍奇金淋巴瘤与健康对照组血清中差异表达的蛋白。方法选择山西省肿瘤医院72例非霍奇金淋巴瘤患者,男性47例,女性25例;年龄11~83岁,中位年龄52.5岁。正常对照组32例,男性17例,女性15例;年龄10~74岁,中位年龄26.5岁,均为健康志愿者。应用SELDI-TOF-MS技术通过WCX-2蛋白芯片检测他们血清中的蛋白质谱,运用Ciphergen Proteinchip 3.0版本的分析软件自动采集数据,结果采用Biomarker Wiz-ard软件分析二组间蛋白质谱中的差异蛋白。结果笔者在非霍奇金淋巴瘤患者的血清中找到了一系列特异表达的蛋白质,其质荷比分别为3193 Da,3398 Da,7974 Da,8564 Da,8693 Da和15934 Da。72例NHL患者与30例正常人的血清中有5个差异明显的潜化标志蛋白,患者血清中高表达且正常人血清低表达的蛋白质的相对分子量为15 934 Da,患者血清中低表达且正常人血清高表达的蛋白质相对分子量为3193 Da,3398 Da,8564 Da,8693 Da。结论质荷比3193 Da,3398 Da,8564 Da,8693 Da和15 934 Da的蛋白质可能是非霍奇金淋巴瘤的生物学标志物。

【Abstract】 Objective To find differential expression protein by using surface-enhanced laser desorption-ionization time-of-flight mass spectrometry(SELDI-TOF-MS) patients with non-Hodgkin Lymphoma(NHL) and healthy controls.Methods Seventy-two cases of NHL,including 47 male and 25 female,ages from 11 to 83(mean 52.5).Thirty-two healthy persons,including 17 male and 15 female,ages from 10 to 74(mean 26.5) were as control.Serums from them were detected by SELDI-TOF-MS(WCX-2 portein Chip and Ciphergen software) was used to identify proteomic features.Results A number of potential candidate proteins,including the peak intensity of 3193 Da,3398 Da,7974 Da,8564 Da,8693 Da and 15 934 Da,were identified in patients with NHL.There were five differentially expressed protein markers between 72 NHL patients and 32 healthy volunteers.The protein marker of 15 938 Da was high level in patients,but low level in healthy control.The protein markers of 3193 Da,3398 Da,8564 Da and 8692 Da weare low level in patients,but high level in healthy volunteers.Conclusion These peaks of 3193 Da,3398 Da,7974Da,8564 Da,8693 Da and 15 934 Da are potential biomarkers for the diagnosis of non-Hodgkin lymphoma(NHL).
