外周血高分辨染色体制备方法研究

【摘要】 目的建立一种简易的人外周血高分辨染色体标本制备方法。方法将30份外周血样本随机分为3组,每组10份:A组采用常规法制备G显带染色体,B组采用过量胸苷(TDR)同步化法制备高分辨染色体,实验组采用低温休克和低浓度的秋水仙胺双重同步处理细胞,再用热蒸气—过氧化氢法制备高分辨染色体。结果实验组与A组相比,细胞分裂指数差异无统计学意义(P>0.05),但实验组获得的早期染色体分裂相数较多,两组比较差异具有统计学意义(P<0.01);与B组相比,两组获得早期染色体分裂相数无差异(P>0.05),但B组的细胞分裂指数较低(P<0.05)。另外,实验组染色体分散较好,G显带清晰。结论该法不需昂贵的试剂,操作简便易于掌握,能获得较多带纹清晰的高分辨染色体,适合于临床检测应用。

【Abstract】 Objective To search a simple method for the preparation of high-resolution peripheral blood chromosome.Methods Thirty periphearal blood samples were randomly divided into three groups of ten.In Group A,a routine method was applied to the preparation of G chromosome banding.In Group B,a synchronization method with excessive TDR was applied to the preparation of high-resolution chromosome.Group C was the experimental group,the combined treatments of low-temperature shock and low concentration colchicine were applied synchronously to deal with cells,then steam and hydrogen peroxide were applied.Results There was no significant difference in cell division index between Group A and the experimental group(P>0.05),but in Group C,the number of early chromosome metaphase was comparatively more and the difference was significant(P<0.01);there was no significant difference in the number of early chromosome metaphase between Group B and the experimental group(P>0.05),but in Group B,the cell division index was lower(P<0.01).In addition,chromosome spread in the experimental group was better and the G banding was clear.Conclusion The method of synchronous treatments of low-temperature and low concentration colchicines followed by steam and hydrogen peroxide for the preparation of high-resolution peripheral blood chromosome is simple,economic and may obtain much more high-resolution chromosomes with clear lines.The method can be used for clinical application.
