动态增强MRI评价内皮抑素对小鼠结肠癌肿瘤血管生成的研究

【摘要】 目的 探讨动态增强MRI评价内皮抑素对小鼠结肠癌肿瘤血管生成的价值。方法 建立小鼠结肠癌皮下移植瘤模型,1周后筛选成瘤均匀的32只随机分成内皮抑素(ES)组和生理盐水(NS)组,分别给予内皮抑素6 mg/(kg·d)和0.2 ml生理盐水皮下注射,连续注射14 d后行MRI动态增强扫描,观察MRI动态增强时间-信号强度曲线(time-signal intensity curve,TIC)形态,并比较首次增强斜率(Sfirst)的差异及其与MVD、VEGF表达间的相关性。结果 ES组TIC呈缓慢上升型,而NS组呈速升缓降型。NS组的Sfirst为(4.92±1.03) units,ES组为(3.17±0.91) unit/s,NS组的Sfirst明显高于ES组(P<0.05)。ES组VEGF表达水平较NS组降低、MVD分布也较稀疏。Sfirst与VEGF及Ⅷ因子的表达均呈明显正相关(P<0.05)。结论 动态增强MRI能较准确评价内皮抑素对小鼠结肠癌肿瘤血管生成的影响。

【Abstract】 Objective To explore the value of dynamic contrast-enhanced MRI in evaluating the angiogenesis of endostatin in colorectal carcinoma.Methods The models of colorectal carcinoma subcutaneously transplantation in mice were established,one week later,32 tumor-bearing mice were sieved out and divided randomly into two groups:group A and group B,treated with intraperitoneal injection of endostatin(ES) 6 mg/(kg·d) and 0.2 ml normal saline(NS) respectively for 14 days.Subsequently,dynamic contrast-enhanced MRI was performed and the shapes of time-signal intensity curve(TIC) were observed.The difference of first slope was compared and the relation of the first slope and MVD and VEGF expression between group A and B,respectively.Results The TIC appeared as a slowly ascending type in group A and rapidly ascending-slowly descending one in group B.The first slopes were(4.92±1.03) unit/s and(3.17±0.91) unit/s in group A and B respectively.There was significant difference between these two groups(P<0.05).The expressions of VEGF and MVD in group A were lower than which in group B.There was positive correlation between first slope and expressions of VEGF and MVD(P<0.05).Conclusion Dynamic contrast-enhanced MRI can evaluate correctly the angiogenesis of endostatin in the models of colorectal carcinoma in mice.
