辛基酚聚氧乙烯醚降解菌的分离鉴定及降解特性研究
Isolation,Identification Study on Octylphenol Polyethoxylate Degrading Strains and its associated Degradation Characteristics
【摘要】 从农田土壤及池塘水中分离得到共2株降解内分泌干扰物辛基酚聚氧乙烯醚(OPEOn)的细菌X-4及X-6.经过对其形态特征、生理生化以及16S rDNA序列分析,菌株X-4初步鉴定为球形芽孢杆菌(Bacillus sphaeri-cus),菌株X-6初步鉴定为嗜水气单胞菌(Aeromonas hydrophila)或点状产气单胞菌点状亚种(Aeromonas puncta-ta)。通过摇瓶实验考察了生长条件对OPEOn降解的影响,得出X-4及X-6降解OPEOn的最佳条件是接种量为1.0%,初始pH值为7.3~7.5,温度为30℃。在该条件下,对菌株X-4及X-6在不同初始浓度OPEOn下的降解反应过程进行动力学分析,该降解反应在低质量浓度(5,20,50,100mg/L)时符合一级反应动力学特征,而在较高底物质量浓度(200mg/L,1000mg/L)下不能用一级反应动力学描述,且2株菌对OPEOn的降解率在初始浓度为20mg/L时达到最大值,分别为X-4:26%,X-6:32%。
【Abstract】 Two strains of octylphenol polyethoxylate(OPEOn)degrading,X-4 and X-6 were separated and obtained from farmland soil(X-4)and pond water(X-6).The strains were identified preliminarily as Bacillus sphaericus for X-4,Aeromonas hydrophila or Aeromonas punctata for X-6 based on analysis in terms of morphological characteristics,physiological and biochemical characterization,as well as sequence analysis of 16S rDNA.Growth environment for the two strains were tested though a designed shaking flasks experiment,The optimum condition indicate that pH remain at 7.3-7.5,,temperature reach 30 ℃,with inoculum size 1.0% for X-4 and 1.5% for X-6,respectively.Under such circumstances,the degradation reaction are consistent with one order kinetic characteristic in the case of lower initial level for strains(5,20,50,100 mg/L),while it failed to follow one order kinetic characteristic.while higher initial sample level present(200 mg/L,1 000 mg/L).Furthermore,when the initial concentration for strains registered 20mg/L,the degradation rate maximized with 26% for X-4 and 32% for X-6,respectively.
