【摘要】 目的探讨慢性粒细胞白血病(Chronic myelogenous leukemia,CML)来源的树突状细胞(Dendritic cell,DC)激活的T淋巴细胞对自体CML细胞的杀伤作用。方法利用rhGM-CSF、rhIL-4和rhTNF-α体外定向诱导CML患者外周血单核细胞(Peripheral blood mononuclear cell,PBMC)生成DC,光学显微镜和透射电镜观察所诱导的DC形态特征;流式细胞术进行表型检测;采用FISH和RT-PCR法检测所诱导的DC的白血病源性;MTT法检测其对自体CML细胞的杀伤作用。结果 CML患者PBMC在体外可诱导生成bcr/abl融合基因阳性的DC,其激活的T淋巴细胞对自体CML细胞的杀伤作用明显强于单独应用T淋巴细胞对CML细胞的杀伤作用。结论 CML患者PBMC能够诱导生成白血病源的DC,该DC能够激活T淋巴细胞对CML细胞的特异性杀伤作用。更多还原

【Abstract】 Objective To investigate the killing effect of chronic myelogenous leukemia(CML)-derived dendritic cells(DCs)-activated T lymphocytes on autologous CML cells.Methods The peripheral blood mononuclear cells(PBMCs) of patients with CML were induced directly in vitro with rhGM-CSF,rhIL-4 and rhTNF-α,and the produced DCs were observed for morphology by light microscopy and transmission electron microscopy,then analyzed for phenotype by flow cytometry,for leukemia derivation by FISH and RT-PCR methods,and for killing effect on autologous CML cells by MTT method.Results The PBMCs of patients with CML induced the production of DCs positive for bcr/abl fusion gene in vitro.Compared with T lymphocytes administered alone,the T lymphocytes activated with the DCs showed significantly killing effect on CML cells.Conclusion The PBMCs of patients with CML induced CML-derived DCs which activated the specific killing effect of T lymphocytes on CML cells.更多还原
