菌紫质的生物素标记和光电响应(英文)

【摘要】 用生物素标记紫膜(purple membrane,PM)中的菌紫质(bacteriorhodopsin,BR)后,紫外-可见吸收光谱测定表明,PM的BR中,视黄醛微环境没有明显变化;时间分辨动力学光谱研究发现,生物素化的PM中,BR保留了光循环的活性和质子泵的功能,但这些功能有所下降。通过双层脂膜(bilayer lipid membrane,BLM)的光电响应测定可知,生物素化的PM中,BR同样具有光电响应活性。在本工作中,我们也在BLM系统上研究了生物素化的BR与抗生物素之间的分子识别,并进行了定向组装,测量结果表明,定向组装的生物素化PM,其光电压值要比非特异性吸附的光电压值大20倍,而其在BLM上的量只比非特异性吸附的PM大2倍。

【Abstract】 The bacteriorhodopsin(BR) in purple membrane(PM) was labeled by biotin,and illustrated by the characteristic UV-VIS absorption spectra.The results showed that microenvironment where retinal existed within BR of biotinylated PM had not been disturbed.Flash kinetic absorption measurements suggest that the activities of photocycle and proton pump of biotinylated PM had been maintained but weakened.It was indicated by the photoelectric response in bilayer lipid membrane(BLM) that biotinylated PM also maintained its photoelectric activity.In this work,we completed the oriented assembly of biotinylated PM by biotin/streptavidin molecular recognition in the BLM system.The photoelectric voltage peak of oriented biotinylated PM in the BLM was 20-fold larger than that of non-specific absorbed PM although the amount of the biotinylated PM on the membrane was about 2-fold more than PM on the membrane.
