伏马毒素B1酶标试纸条检测方法

【摘要】 以膜为载体,开发一种直接竞争酶标试纸条——dipstick,用于快速检测谷物类样品中的伏马毒素B(1FB1)并对试纸条的检测参数进行优化。该浸蘸式酶标试纸条对FB1的最低检测限为10μg/L。其检测过程简单,所需时间短,大约20 min即可完成。结果通过目视直接辨别,可以在1 000μg/kg毒素水平上给出"是"或"否"的回答。HPLC验证结果表明dipstick方法可以用于谷物中FB1的快速定性和半定量检测。

【Abstract】 A test strip method-dipstick based on direct competitive enzyme-linked immunosorbent assay(ELISA) for rapid detection of fumonisin B1(FB1) in corn samples was developed.Assay conditions,including concentrations of antibody and enzyme conjugate,coating buffers and so on,were optimized.The optimized dipstick immunoassay allowed FB1 determination with a limit of detection of 10 μg/L.The assay can rapidly detecte FB1 within 20 min and the result was capable of investigation just through eyes on the FB1 concentration of 1 000 μg/kg.Good correlation with spiked food samples was obtained using HPLC method.The developed dipstick immunoassay was capable of being applied for the online monitoring of FB1 in food samples.
