反相高效液相色谱法测定紫山药中薯蓣皂苷的含量

【摘要】 建立薯蓣属中紫山药中薯蓣皂苷含量测定方法,考察不同产地紫山药中薯蓣皂苷的含量,并与该属中其他山药进行对比。样品经冷冻干燥后研磨成粉末,然后经70%甲醇回流提取1h,采用HPLC法进行测定。色谱柱:DiamonsilTM(钻石)C18柱(250mm×4.6mm,5μm),流动相:甲醇-水(84∶16),流速:1.0mL·min-1,检测波长:206nm,柱温:30℃。结果表明,薯蓣皂苷与其他成分分离良好,线性范围0.65~65μg·mL-1(r=0.9999),回收率100.6%,不同产地山药中薯蓣皂苷含量差异较大。该方法简便、准确、重现性好,可用于山药及其同属其他植物中薯蓣皂苷的质量控制。

【Abstract】 To develop a RP-HPLC method for the determination of dioscin in purple Dioscorea alata Linn.After being extracted by refluxing with 70% MeOH for 1 hour,samples were achieved and separated by DiamonsilTM C18 column(250mm×4.6mm,5μm)with methanol-water(84∶16)as mobile phase.The flow rate was 1mL·min-1.The column temperature was 30℃.The UV detector was set at 206nm.Dioscin was well-separated.The linear calibration curve was obtained in the concentration range of 0.65~65μg·mL-1(r=0.9999),the average recovery of dioscin was 100.6%.The contents of dioscin in purple Dioscorea alata Linn.of different habitats was in larger difference.The method was simple,accurate and reproducible.It was also proposed to be used for the quality control of dioscin in Dioscorea opposite Thunb and Dioscorea alata Linn.
