白腐真菌产木聚糖酶发酵条件的优化及酶学性质研究
Study on the optimization of xylanase fermentation conditions by white rot fungi and the enzymological properties
【摘要】 通过正交实验对白腐真菌Pleurotus ostreatus SYJ042产酶条件进行了优化,研究了该酶的酶学性质。结果表明:最适发酵条件为:碳源为5%的麸皮和玉米芯(1:1),氮源为0.5%的蛋白胨,接种量为每50mL发酵液接4块菌丝块,发酵时间为8.5d。该酶的最适温度是50℃,最适pH为5.4;Zn2+、Ca2+对酶活性影响较小;Ag+、MnO2-4影响较大。
【Abstract】 By the orthogonal test,the optimal fermented condition of xylanase secretion by white rot fungi was obtained:2.5% wheat bran+2.5% corn cob,0.5% peptone,4 pieces per 50mL liquid volume,8.5d.By the experimental method in the thesis,some properties of the xylanase were obtained:the optimal temperature was at 50℃.The optimal pH was 5.4.Ag+ and MnO2-4 evidently impacted on the xylanase activity.
