银杏叶提取物EGb761对老化红细胞的保护作用

【摘要】 目的观察银杏叶提取物EGb761对红细胞老化的影响,探讨其延缓红细胞衰老的机制。方法采用37℃温箱孵育模拟红细胞的自然衰老过程,比较单纯衰老组、不同浓度EGb761保护组(EGb761的浓度分别0.002、0.010、0.050、0.250 g/L)和正常红细胞组(正常对照组)老化红细胞丙二醛(MDA)含量、红细胞超氧化物歧化酶(SOD)活性及红细胞膜骨架结构。结果与正常对照组比较,单纯衰老组红细胞MDA含量明显增高,SOD活性明显下降,细胞膜骨架结构破坏;不同浓度EGb761保护组衰老红细胞的MDA含量明显降低,SOD活性明显升高,差异均有极显著性(F=193.95、361.41,q=6.08~113.29,P<0.05、0.01),细胞膜骨架结构破坏也减轻,且随着其浓度增高作用逐渐增强。结论 EGb761能在细胞膜水平上延缓氧自由基引发的脂质过氧化作用导致的红细胞老化过程,保护红细胞及其膜骨架结构。

【Abstract】 Objective To observe the effects of Ginkgo biloba extracts(EGb761) on erythrocyte aging,and explore its mechanism of delaying the senescence of erythrocytes. Methods Incubating erythrocytes in 37 ℃ box to simulate erythrocyteaging process,they were then divided into merely-aging group,protection group of EGb761 of different concentrations,i.e.0.002,0.010,0.050 and 0.250 g/L,and normal control group.The levels of product of erythrocyte lipid peroxidation-malondialdehyde(MDA),the activity of superoxide dismutase(SOD),and membrane skeleton structure(MSS) in erythrocyte were compared with that of the normal control. Results Compared with the control group,the levels of MDA in the merely-aging group significantly elevated,the activity of SOD decreased,the MSS of erythrocytes was destroyed;in the different protection groups,the MDA levels in aged erythrocytes declined,SOD enzyme elevated(F=193.95,361.41;q=6.08-113.29;P<0.05,0.01),the destruction of MSS lessened,the effect became stronger with the increase of the concentration. Conclusion EGb761 can,at the membrane level,delay the erythrocyte lipid peroxidation that induces the process of erythrocyte aging,protecting erythrocytes and MSS.
