矽肺大鼠血清硅元素水平及Clara细胞蛋白的表达和意义
Study on levels of silicon and CC16 in serum of rats exposed to silica dust
【摘要】 目的探讨不同染尘剂量大鼠矽肺模型血清硅元素水平的变化情况,并与反映肺内早期炎症变化的细胞因子血清克拉拉细胞蛋白(Clara cell protein,CC16)进行比较,以弄清它们在矽肺发病中的意义和价值。方法雄性Wistar大鼠分为28组,按不同染尘剂量(10 mg/ml、30 mg/ml、50 mg/ml、70 mg/ml)染尘,而后在不同时间点(染尘后1 d、3 d、5 d、7 d、14 d、21 d和28 d)处死动物,采取血液及肺组织,用电感耦合等离子体质谱仪(ICP-MS)检测血清中硅元素含量,酶联免疫吸附测定法(ELISA法)测定大鼠血清中CC16含量。结果随着染尘剂量增加和染尘时间延长,大鼠血清硅元素的水平呈现逐渐增高的趋势;CC16含量于染尘后即开始降低,第7天开始与对照组相比差异有统计学意义(P<0.05)。并随着染尘时间延长和剂量增加,其含量亦不断减少,但血清硅元素的变化不论是从时间上还是从剂量上比较,均早于CC16。结论矽肺模型大鼠血清硅元素水平在早期低剂量染尘时即明显升高,且早于CC16的变化,二者的变化具有剂量和时间依赖性,提示血清硅元素、CC16水平变化可能为矽肺的早期诊断提供生物学依据,具有一定的临床价值。
【Abstract】 Objective To explore the change of serum silicon levels in rats with different exposure doses,and compare to the changes of serum Clara cell protein(CC16)which is an early biomarker of inflammation in lung,and clarify their roles in pathogenesis of silicosis.Methods Male rats were randomly divided into 28 groups according to their exposure doses to silica(10,30,50 or 70 mg/ml)and their exposure time(1 d,3 d,5 d,7 d,14 d,21 d and 28 d).The serum levels of silicon were determined by inductively coupled plasma mass spectrometry(ICP-MS),the serum levels of CC16 were examined by ELISA method.Meantime,the pathological changes in lung were also observed using hematoxylin-eosin staining.Results The silicon level in serum was gradually increased in a dose-and time-dependent manner,however,the CC16 level in serum was dose-dependently decreased but later than that of silicon level.Conclusions The results showed that the changes of serum levels of silicon and CC16 in rats appear quite early after exposure to silica dust,especially serum silicon level,and the changes were dose-and time-dependent.It is suggested that these indices mentioned above might provide some earlier evidence of silicosis,therefore should have certain clinical significance.
