食管鳞癌组织PTTG和bFGF的表达与临床病理特征的相关性研究

【摘要】 目的探讨食管鳞癌组织中垂体瘤转化基因(PTTG)mRNA及其蛋白和碱性成纤维细胞生长因子(bFGF)蛋白的表达及相互关系,以及其与肿瘤临床病理指标及预后的关系。方法采用RT-PCR技术检测40例食管癌及相应的40例癌旁组织中PTTG mRNA的表达,同时采用免疫组织化学SP法检测相应标本中PTTG和bFGF蛋白的表达。结果在食管癌组织中PTTG基因呈过度表达,食管癌组织中PTTG mRNA的平均表达水平(0.71±0.08)显著高于相应的癌旁正常组织(0.32±0.06),两组比较,差异有统计学意义(P<0.01)。免疫组化结果显示,PTTG蛋白在食管癌组织中的表达(34/40)明显高于在癌旁组织中的阳性表达(9/40)(P<0.01)。其阳性率高低,与TNM分期和分化程度有关。食管癌组织中,bFGF蛋白表达阳性、阴性标本中,PTTG mRNA表达水平分别为0.74±0.03和0.59±0.02。结论 PTTG基因和bFGF基因的异常表达可对食管癌的发生和发展起重要作用,它们可能为食管癌的早期诊断和基因治疗提供新的靶点。

【Abstract】 Objective To explore the expressions of mRNA and its protein of pituitary tumor-transforming gene(PTTG)in esophageal squamous cell carcinoma and the expression of basic fibroblast growth factor(bFGF)as well as their correlation,and the relationship between their expressions and tumor clinical pathological indexes and prognosis.Methods RT-PCR technology was used to detect the expression of PTTG mRNA in 40 cases of esophageal carcinoma and in 40 cases of corresponding pericarcinomatous tissues.SP method of immunohistochemical technology(IHC)was adopted to detect the expression of PTTG and bFGF proteins in corresponding samples.Results PTTG was over-expressed in esophageal carcinoma.The mean expression level of PTTG mRNA in esophageal carcinoma(0.71±0.08 respectively)were higher significantly than that in corresponding pericarcinomatous tissues(0.32±0.06 respectively).The differences in those were significant statistically in comparison of two groups(P<0.01).IHC results showed that the expression of PTTG protein in esophageal carcinoma(34/40)was apparently higher than that in the positive expression in pericarcinomatous tissues(9/40)(P<0.01).The level of its positive rate was relevant with TNM staging and differentiation.In esophageal carcinoma and in the samples with bFGF protein positive and negative expressions,the expressions of PTTG mRNA were 0.74±0.03 and 0.59±0.02 respectively.Conclusion The abnormal expressions of PTTG and bFGF may play the important role in the occurrence and development of esophageal carcinoma.They may provide the new targets for the early diagnosis and genetic treatment of esophageal carcinoma.
