PAR4在背根神经节感觉神经元的表达及与TRPV1和CGRP的共存

【摘要】 目的:研究蛋白酶活化受体4(PAR4)在小鼠背根神经节(DRG)感觉神经元的表达,及与瞬时受体电位香草酸亚型1(TRPV1)和降钙素基因相关肽(CGRP)的共存,为探讨PAR4在感觉神经伤害性刺激信号传导中的作用提供形态学依据。方法:免疫荧光组织化学双标方法结合激光共聚焦显微镜技术。结果:DRG内大量的感觉神经元表达PAR4。PAR4阳性胞体多为中、小型神经元,并可见部分阳性神经纤维。免疫荧光双标显示许多PAR4阳性神经元表达TRPV1或与CGRP共存。几乎所有的TRPV1阳性神经元均表达PAR4,大部分CGRP标记神经元呈PAR4阳性,另外还可见到许多CGRP/TRPV1双标神经元。结论:小鼠DRG初级感觉神经元广泛的表达PAR4,该受体可能通过影响TRPV1和CGRP参与伤害性刺激的调节。

【Abstract】 Objective: To study the expression of the protease-activated receptor-4(PAR4) and its co-localization with transient receptor potential vanilloid type 1(TRPV1) or calcitonin gene-related peptide(CGRP) in the sensory neurons of the mouse dorsal root ganglia(DRG),and provide the morphological evidence for exploring the role of PAR4 in the signal transduction of nociceptive stimuli in primary afferents.Methods: Double labeling fluorescent immunohistochemistry combined with laser scanning confocal microscope techniques were used.Results: The vast majority of neurons throughout DRG displayed PAR4 expression,and most of PAR4 immunoreactive neuronal cell bodies were middle and small in size.Some PAR4 immunoreactive fibers were observed.Double-labeling fluorescent histochemistry with PAR4 and TRPV1 or CGRP showed that most of PAR4 immunoreactive neurons were co-expression with TRPV1 or co-localization with CGRP in the sensory neurons of the DRG.On the other hand,almost all of the TRPV1 immunoreactive neurons expressed PAR4,and the majority of CGRP immunopositive neurons co-contained with PAR4 immunoreactivity.Also,most CGRP immunoreactive DRG neurons co-expressed TRPV1.Conclusion: The present findings demonstrate that PAR4 is extensively expressed in primary afferent neurons of the mouse DRG,where they might involve in nociceptive responses by through the modulation TRPV1 and CGRP.
