尿感方抗尿道致病性大肠杆菌的实验研究

【摘要】 目的:观察尿感方对尿道致病性大肠杆菌(UEC)在人尿道上皮细胞黏附效应的影响。方法:体外实验以营养肉汤组作空白对照,观察经三金片、蔓越莓、不同浓度的尿感方等药物处理后人尿道上皮细胞上UEC黏附数量的变化。体内实验比较经正常尿液及口服尿感方后4 h尿液、次日晨尿处理后的人尿道上皮细胞上UEC黏附数量的变化。结果:①体外实验:经不同药物处理后的各用药组其UEC黏附于尿道上皮细胞上的细菌数较未经药物处理的营养肉汤组明显减少(P<0.01);三金片、蔓越莓组与同等浓度的尿感方组比较,其黏附在尿道上皮细胞上的UEC细菌数无明显差异(P>0.05);三种不同浓度的尿感方组比较,尿感方浓度越高,抗黏附效果越好。②体内实验:经口服尿感方后4 h尿及次日晨尿处理过的UEC黏附于尿道上皮细胞上的数目较经服药前的正常尿液处理的UEC黏附数目明显减少(P<0.05,P<0.01)。结论:尿感方具有良好的体外、体内抗UEC黏附的作用,且药物浓度越高,抗黏附效果越好。尿感方阻止UEC在人尿道上皮细胞的黏附可能是其治疗尿路感染的作用机制之一。

【Abstract】 Objective:To observe the effects of "Niaogan Formula"(NGF) on adherence of Uro-pathogenic Escherichia Coli(UEC) to human uro-epithetial cell(HUEC).Methods:In vitro:compared with the blank control group(broth),the number of the UEC adhered to HUEC was calculated after management of "Sanjin Tablet",Cranberry Juice Cocktail,and NGF of different concentrations.In vivo:the number of the UEC adhered to HUEC was calculated after management of the normal urine,the 4-hour and next-morning urine after taking NGF.Results:①In vitro:the number of the UEC adhered to HUEC significantly decreased after management of different drugs compared with the blank control group(P<0.01);there were no obvious differences in the number of the UEC adhered to HUEC between "Sanjin Tablet",Cranberry Juice Cocktail and NGF of the same concentration(P>0.05);with the higher concentration of NGF,the effect of anti-adhesion was better.②In vivo:the number of the UEC adhered to HUEC significantly decreased after management of the 4-hour and next-morning urine after taking NGF compared with the normal urine(P<0.05,P<0.01).Conclusion:NGF plays a good role in prohibiting UEC from adhering to HUEC in vivo and vitro,and with the higher concentration of NGF,the effect of anti-adhesion is better.NGF inhibiting UEC from adhering to HUEC may be one of its mechanisms against urinary infection.
