【摘要】 目的研究低分子硫酸皮肤素(LMWDS)的抗血栓作用及其时效、量效关系,确定其半数有效量(ED50)。方法健康家兔sc低分子硫酸肝素(LMWH)、硫酸皮肤素(DS)和不同剂量的LMWDS,取血浆测定LMWDS对家兔凝血系统、血小板和纤溶系统的各指标的影响;采用改良的Chandler法测定时效、量效关系。结果 LMWDS能够剂量依赖性地增强肝素辅因子和调节组织型纤溶酶原激活物(tPA)的活性并抑制纤维蛋白原(Fg)的产生,但是对凝血酶原时间、凝血酶时间、活化凝血酶原时间和血小板聚集没有影响。LMWDS在给药后2 h血栓抑制率最强,在8 h时又表现出较强的抗血栓活性,然后消除。健康家兔sc LMWDS的ED50为5.337 mg/kg。结论 LMWDS具有良好的抗血栓作用,并且具有剂量依赖性和时效性。更多还原

【Abstract】 Purpose To study on antithrombotic effect,the time-effect and dose-effect relationship of LMWDS,and to confirm the ED50 at the same time.Methods In vitro,PPP and PRP were collected from rabbits and antithrombotic effect was determined.The improved Chandler method was used to determine the time-effect and dose-effect of LMWDS.Results The studies on the antithrombotic effect of LMWDS showed that it dose dependently enhanced the activity of HCII and tPA,inhibited Fg production,but it had no effect on PT,TT,AT III and platelet aggregation.The study on the time-effect relationship of LMWDS showed that its thrombotic inhibition effect reached its maximum at 2 h after sc and a second peak of thrombotic inhibition appeared at 8 h.The dose-effect relationship study showed that sc LMWDS was dose dependent.The ED50 value of sc LMWDS was 5.337 mg/kg.Conclusion LMWDS have excellent antithrombotic effect,which is dose-dependent and chronergic.更多还原
