大叶杨花粉离体萌发和贮藏条件的研究

【摘要】 以15~20 a生大叶杨(Populus lasiocarpa Oliv.)花粉为试材,研究了不同培养基对其萌发的作用,初步探讨了贮藏温度对花粉生活力的影响。结果表明,花粉在25℃下培养24 h萌发率趋于稳定;培养基中添加蔗糖和H3BO3可使花粉萌发率显著提高;随蔗糖和H3BO3浓度升高,萌发率呈现先升高再降低的趋势,在5%蔗糖+50 mg/L硼酸的培养基上达到最高,为56.33%;培养基中添加50~200 mg/L GA3对大叶杨花粉的萌发具有明显的抑制作用;花粉自然干燥后贮藏于-20℃条件下较为适宜。本研究结果对利用大叶杨优良资源培育优良山地杨树新品种,进一步扩大杨树种植区域,解决平原地区土地资源紧张等问题具有重要意义。

【Abstract】 Effects of different medium and storage temperature on pollen germination of 15~20 year-old Populus lasiocarpa Oliv.were studied.The results indicated that pollen germination rate tended to be stable after culturing for 24 h under 25 ℃.Pollen germination rate increased greatly when sucrose and boric acid was supplemented in the medium.As the concentrations of sucrose and boric acid increased,the pollen germination rate ascended firstly,and then descended.The highest pollen germination rate(56.33%) was observed on the medium with 5% sucrose and 50 mg/L H3BO3.50~200 mg/L GA3 inhibited pollen germination significantly.The optimum pollen storage condition was under-20 ℃ after natural drying.These results are of significance for breeding elite cultivars in mountain areas utilizing P.lasiocarpa as excellent resources,broadening Populus planting regions and resolving the problem of land shortage.
