延胡索与不同中药配伍对疼痛模型小鼠血清和脑组织NO含量昼夜节律的影响
Effect of Corydalis Tuber Combined With Different TCM on Circadian Rhythm of Nitric Oxide Content in Serum and Brain Tissue of Pain Model Mice
【摘要】 目的:研究延胡索与不同中药配伍对疼痛模型小鼠血清、脑组织中一氧化氮(NO)含量昼夜节律的影响。方法:280只小鼠随机分为正常组、模型组、单味延胡索组、祛瘀止痛组、温阳止痛组、补虚止痛组、清热止痛组7组;每组又分为子时、卯时、午时、酉时4个时辰药物处理组,共28组,每组10只。于末次给药后30 min,各组小鼠按0.1 mL/10 g体重,腹腔注射0.6%冰醋酸致痛。采用硝酸还原酶法,酶联免疫检测仪检测小鼠血清、脑组织中NO的含量。结果:与模型组比较,单味延胡索组、补虚止痛组和清热止痛组中小鼠血清、脑组织中NO含量在各时辰均表现为一定程度的下降,尤以暗期的表现明显(P<0.05);而温阳止痛组和祛瘀止痛组小鼠血清、脑组织中NO含量暗期表达显著升高(P<0.05)。结论:该实验为临床痛证的择时治疗提供了相关的实验的和理论的依据。
【Abstract】 Objective:To investigate the effect of corydalis tuber combined with different traditional Chinese medicine on circadian rhythm of nitric oxide content in serum and brain tissue of pain model mice.Methods:280 mice were randomly divided into control group,model group,corydalis tuber group,group of removing blood stasis to alleviate pain,group of warming yang to alleviate pain,group of expelling heat to alleviate pain,group of reinforcing deficiency to alleviate pain.Every group contains four subgroups which were administrated at mao shi(05∶00),wu shi(11∶00),you shi(17∶00)and zi shi(23∶00)respectively.30 minutes after the last administration,mice was injected with glacial acetic acid(0.1 mL /10 g body weight).Then nitric oxide(NO)content in serum and brain tissue of mice was detected by nitrate reductase method.Results:Compared with model group,NO content in serum and brain tissue of mice was decreased in dark period in Corydalis tuber group,group of expelling heat to alleviate pain,group of reinforcing deficiency to alleviate pain(P<0.05),but it was increased in dark period in group of removing blood stasis to alleviate pain,group of warming yang to alleviate pain(P<0.05).Conclusion:This experiment provide experimental and theoretical proof for time-selecting therapy of pain syndrome.
