人染色体端粒酶基因的扩增率与宫颈癌前病变和宫颈癌的研究

【摘要】 目的:探讨人染色体端粒酶基因(hTERC)扩增率与宫颈癌前病变及宫颈癌的关系。方法:应用荧光原位杂交技术(FISH)检测宫颈上皮内瘤变(CIN)组108例(CINⅠ39例、CINⅡ37例、CINⅢ32例),宫颈癌组24例和正常对照组33例,共165例宫颈脱落细胞中hTERC基因的表达情况。结果:CINⅡ、CINⅢ和宫颈癌hTERC基因的扩增率,分别为64.86%、75.00%和91.67%,明显高于CINⅠ(15.38%)和正常对照组(6.06%),差异有统计学意义(P<0.05);CINⅠ组与正常对照组hTERC基因扩增率比较,差异无统计学意义(P>0.05)。结论:hTERC基因的扩增率在各级宫颈病变均有一定的比例。hTERC基因的扩增率随着宫颈病变分级呈逐渐上升趋势,且hTERC基因在高度病变中的扩增率,明显高于低度病变。

【Abstract】 Objective: To investigate the correlation between amplification rate of Human Telomerase RNA Component(hTERC) gene and cervical precancerous lesion and cervical cancer.Methods: The expressions of hTERC gene in 165 cases with exfoliated cervical cells were detected by the fluorescence in situ hybridization (FISH),which including 108 cases with CIN (39 CINⅠ,37 CIN Ⅱ,32 CIN Ⅲ),24 cases with cervical cancer,33 cases with normal controls.Results: Positive expression rates of the hTERC gene in CIN Ⅱ,CIN Ⅲ and cervical cancer were 64.86% ,75.00% and 91.67%,respectively which was obviously higher than that in CIN Ⅰ (15.38%) and normal controls (6.06%).There was no statistical difference between CIN Ⅰ and normal control about hTERC amplification rate (P>0.05).Conclutions: hTERC gene amplification can be detected in each grade of CIN and the positive rate is increased with the grade upward.The positive rates of hTERC gene in high-grade lesions are significantly higher than that in low-grade lesions.
